用薄层扫描法测定油茶粕蛋白水解物的多肽

摘  要:
为了研究薄层扫描法测定油茶粕固态发酵蛋白水解物中多肽的含量的可行性,在确定展开剂及描述波长的基础上,测定了该方法的重复性、加样回收率,并与高效液相色谱法进行了含量的的对比。结果表明:以谷胱甘肽为标准品、茚三酮为显色剂,样品液中多肽的含量为1.82 mg/mL,而原料中多肽含量只有0.89mg/mL,该方法重复性试验标准偏差为0.03%;回收试验的平均回收率为106.94%,标准偏差为0.058%,重现性和加样回收率等均符合含量测定要求。该法测定的含量与反向高效液相色谱法相比无显著性差异(P>0.05)。薄层扫描法适合于在快速检测多肽中应用。

摘  要:
In order to research feasibility of determining polypeptide content in protolysate of Camellia oleifera cake by TCL-scanning,based on determination of developing agent and wavelength,repeatability and recovery of the method were tested,and the result was compared with that by HPLC.The results show that polypeptide content is 1.82 mg/mL in sample solution but 0.89 mg/mL in raw material when using glutathione as standard and monohydrate as chromogenic agent,and standard deviation of the method is 0.03%.Average recovery is 106.94%,standard deviation is 0.058%,and repeatability and recovery meet the demand of content determination.The content determined has no significant differences compared with that by HPLC(P>0.05).The method can be used in fast determination for peptide production.
