摘 要:
以生物降解材料乳酸乙醇酸共聚物 (PL GA,L A- GA 85∶ 1 5 )为载体 ,采用复乳 -液中蒸发法制备含生长激素释放肽 - 6的乳酸乙醇酸共聚物微球 ,并通过体外药物释放试验评价载药微球的释药特征。结果得到收率、粒径大小和分布及含药量均满意的微球 ,其中包封率为 1 0 0 %,平均体积径为 3 .4 5μm ,收率为 79%。在 3 7℃、p H 7.0的生理等渗缓冲液中 ,首日释药 1 9.1 9%,其后以平均日释药 3 .2 3 %的速度释药 2 5 d。小鼠肌肉注射微球 3 0 d后 ,累积增重比注射生长激素释放肽 - 6、生理盐水分别高 2 2 .5 6 %(P<0 .0 5 )和 5 3 .1 7%(P<0 .0 1 )。结果表明 ,生长激素释放肽 - 6乳酸乙醇酸共聚物微球有望发展成为新型长效制剂

摘 要:
Sustained-release GHRP-6 microspheres were prepared by double emulsion-in liquid evaporation process,using biodegradable polylactic-co-glycolicacid as carrier and the drug release test of the microspheres was detected in vitro. The microspheres with good yield,suitable sized and satisfied drug loading was obtained,and significant GHRP-6 sustained was shown in the test of drug release in vitro. The entrapment efficiency was 100%,mean particle size 3.45 μm and the yield above 79%.In pH7.0 physiological isotonic buffer solution,after 19.19% of GHRP-6 was released first day,the microspheres released at rate of 3.23% per day for more than 25 d(r=0.877).After 30 days,accumulative increased body weights on the group injected with GHRP-6 microspheres was significantly higher 22.56%(P<0.05) and 53.71%(P<0.01) than those group injected with GHRP-6 and saline respectively;The conclusion is that the sustained-release GHRP-6 microspheres has been developed successfully as a new long acting GHRP-6 formulation.
