白洗猪线粒体DNA控制区分子遗传变异分析

摘  要:
对濒临灭绝贵州地方猪种白洗猪线粒体DNA控制区进行分子遗传学研究,为其科学保种和合理开发利用提供科学依据。PCR扩增线粒体DNA控制区全序列,直接测序分析。结果表明:白洗猪线粒体DNA控制区全长1 254~1 314 bp,可以分为3个区;按中间重复区序列是否发生变异可以分为A、B 2种类型;聚类分析和单倍型遗传距离分析显示,各类型分布频率很不均衡,遗传多样性呈降低趋势。结果说明,贵州白洗猪可能有2个母系起源,该物种已经濒临灭绝,单倍型多样度较丰富,但是遗传多样性正在逐步丧失,亟需进行科学的保护。

摘  要:
In order to provide a scientific basis on breed conservation and reasonable exploitation and utilization of the Baixi pig, an endangered indigenous pig breed in Guizhou, the mitochondrial DNA control region of it was studied with molecular genetic methods. The complete sequence of the mitochondrial DNA control region was amplified by PCR and analysis were carried out after sequencing. The result showed the complete sequence of the mitochondrial DNA control region is 1254~1314 bp, and it could be divided into three distinct regions. The Baixi pig could be divided into A and B type by judging whether there exist mutations in the repeated sequence area. The results of cluster analysis and haploid genetic distance analysis revealed that the distribution frequency of each type was very uneven and the genetic diversity showed a decreased tendency. In condasion there may be two maternal origins for the Baixi pigs of Guizhou, and the species is on the verge of extinction. Though the haplotype diversity is relatively rich, the genetic diversity of this species is gradually lost, and urgent scientific protection is needed.
