傅里叶红外光谱法测定硫酸酯化葛仙米多糖的取代度

摘  要:
研究以葛仙米多糖合成不同取代度的葛仙米多糖硫酸酯为标样,氯化钡-明胶比浊法测定标样的取代度,傅里叶里红外光谱作为工具测定这一系列产品取代度的方法。吸光度用基线法得到,对所有不同探针谱带、参比谱带和基线作图法的组合进行比较,并通过探针谱带和参比谱带吸光度比值A探针/A参比与取代度的比值的平均相对偏差评价每种组合的优劣。结果表明:对硫酸酯化葛仙米多糖最合适的吸光度比值A探针/A参比为H1261BL1/H1418BL1,结果与传统方法所测得的硫酸酯化取代度相关系数达到0.993。

摘  要:
Sulfated Nostoc sphaeroides Kützing polysaccharides with different degrees of substitution(DS) were used as reference standards for DS determination by a barium chloride-gelatin turbidimetric method.FTIR spectroscopy was used for instrumental determination of DS.The baseline absorbance was recorded.Different combinations of probe band,reference band and baseline were comparatively evaluated based on the average relative deviation of ratio of Aprobe/Areference to DS.H1261BL1/H1418BL1 was found to be optimal for DS calculation with a correlation coefficient of 0.993 in comparison with DS values determined by the traditional method.
