高致病性猪繁殖与呼吸综合征病毒微滴式数字PCR检测方法的建立及初步应用

摘  要:
根据高致病性猪繁殖与呼吸综合征病毒的非结构蛋白(NSP2)基因序列,利用Primer Premier 5.0软件设计合成NSP2特异性的引物和TaqMan探针,建立了高致病性猪繁殖与呼吸综合征病毒的微滴式数字PCR(Droplet digital PCR, ddPCR)检测方法。结果显示,ddPCR最佳反应的引物和探针浓度分别为900 nM和250 nM;特异性和重复性检测结果表明,ddPCR具有良好的特异性和较高的重复性;灵敏度检测结果表明,ddPCR的检测灵敏度比荧光定量PCR高10倍;ddPCR的检测结果与标准品拷贝数的相关系数为R~2=0.999 1,表明该方法检测结果具有良好的线性关系和可信度;用建立好的ddPCR方法对10份临床样品的高致病性PRRSV进行了拷贝数定量。

摘  要:
The primers and probe were designed for Highly Pathogenic Porcine Reproductive and Respiratory Syndrome Virus droplet digital PCR(ddPCR) by using Primer Express 5.0 Analysis Software based on the nucleotide sequence of NSP2.The results showed that the primers and probe concentrations of ddPCR were optimized at 900 nM and 250 nM respectively. The specificity and reproducibility test showed that ddPCR method had advantages on specificity and reproducibility. The sensitivity for ddPCR assay was advantageous over qPCR, which was 10 times higher than that of the qPCR. The liner correlation coefficient(R2) was found to be 0.9991, suggesting that the ddPCR method has high reliability. This method could be used effectively to quantify highly pathogenic PRRSV in ten clinical samples.
