圆齿野鸦椿种子外植体的快繁体系

摘  要:
以圆齿野鸦椿的成熟种子为外植体,研究组培繁殖育苗技术,以筛选最适的培养基配方.结果表明:(1)激素种类和含量对种子无菌株系建立的影响不同,BA含量为0.5-2.5 mg.L-1时,种子的成活率随BA含量的升高而下降,而NAA对其影响不大,种子无菌株系建立的最佳培养基为MS+0.5 mg.L-1BA+0.3 mg.L-1NAA+7.5 g.L-1琼脂+20 g.L-1蔗糖;(2)在细胞分裂素含量相同的情况下,芽的增殖系数随着生长素含量的增加呈先增大后减小的趋势,在MS+1.0 mg.L-1BA+0.5 mg.L-1NAA+1.0 mg.L-1IAA+7.5 g.L-1琼脂+20 g.L-1蔗糖培育基上,发芽率达50%,增殖系数为3.70,芽饱满健壮;(3)细胞分裂素和生长素适当的配比是芽增殖的关键,最佳培养基为MS+2.0 mg.L-1BA+0.3 mg.L-1NAA+1.0 mg.L-1IAA+7.5 g.L-1琼脂+20 g.L-1蔗糖,增殖倍数达5.0;(4)一定含量的NAA有利于诱导长根,而IBA则不同,培养基中NAA的含量为0.1 mg.L-1,IBA的含量为0.5 mg.L-1时,生根率达95%,根长为3.51 cm.

摘  要:
This experiment studied the seedling techniques of tissue culture,which used the mature seeds of Eascaphis konlshli Hayata as explants,selecting out the optimum medium formula.(1) Establishment of aseptic strain: the concentration of BA was 0.5-2.5 mg·L-1,the survival rate declined with the rising of concentration,but it was insignificantly influencd by NAA,the most suitable medium was MS+0.5 mg·L-1 BA+0.3 mg·L-1 NAA+7.5 g·L-1 agar+20 g·L-1 sucrose;(2) Bud induced: under the condition of the same cell mitogen concentration,proliferation coefficient tended to firstly increase and then decrease following the increase of auxin concentration,MS+1.0 mg·L-1 BA+0.5 mg·L-1 NAA+1.0 mg·L-1 IAA+7.5 g·L-1 agar+20 g·L-1 sucrose was the most suitable bud inducing medium,the germination percentage was 50%,the proliferation coefficient was 3.70 and the bud was in perfect condition;(3) Shoot proliferation: the key of proliferation was the suitable matching percentage of cytokinin and auxin,MS+2.0 mg·L-1 BA+0.3 mg·L-1 NAA+1.0 mg·L1 IAA+7.5 g·L-1 agar +20 g·L-1 sucrose was the best shoot proliferation medium,and the proliferation multiple would be 5.0;(4) Rooting: the particular concentration of NAA was good for inducing long root while IBA not,using hormone combination(0.1 mg·L-1 NAA+0.5 mg·L-1 IBA),the rooting rate would be 95% and the average length would be 3.51 cm.
