间接竞争ELISA方法快速检测猪肉组织中的氯丙嗪

摘  要:
[目的]本研究旨在建立一种简单、快速检测猪肉中氯丙嗪(CPZ)残留量的方法。[方法]以CPZ-牛血清白蛋白(BSA)为包被抗原,自制鼠抗CPZ单克隆抗体,在此基础上建立可用于定量检测猪肉中CPZ含量的间接竞争酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA),同时优化检测条件并对该方法进行评价。[结果]采用自行制备的特异性鼠抗CPZ单克隆抗体,建立间接竞争ELISA检测方法,优化得到最佳反应条件:抗原包被稀释倍数为1∶10 000,单克隆抗体浓度为1μg·mL~(-1),酶标二抗稀释倍数为1∶1 000,最低检测限为0.51 ng·mL~(-1),线性检测范围为1.37~111.11 ng·mL~(-1)。线性回归直线方程为y=-1.252 6x+2.853 2,相关系数(R~2)为0.986 6,总变异系数(CV)为6.97%,空白猪肉添加回收率为96.90%~118.79%。[结论]建立的间接竞争ELISA方法特异性强,灵敏度、精密度、准确度和重现性良好,可用于猪肉组织中CPZ残留的快速检测。

摘  要:
[Objectives]The aim of this study was to establish a simple and rapid method for the determination of chlorpromazine( CPZ) residues in pork. [Methods]CPZ-bovine serum albumin( BSA) was used as coating antigen,and mouse anti-CPZ monoclonal antibody was prepared. Based on this,an indirect competitive enzyme-linked immunosorbent assay( ELISA) for quantitative detection of CPZ in pork was established,while the detection conditions was optimized and the performance of the method was evaluated.[Results]The indirect competitive ELISA method was established using specific monoclonal antibodies. The optimal reaction conditions were optimized: the dilution multiple of antigen coating was 1 ∶ 10 000,the dilution concentration of monoclonal antibody was 1 μg·m L~(-1),the dilution multiple of enzyme labeled second antibody was 1 ∶1 000,the limited detection was 0.51 ng·m L~(-1),a linear detection range was 1.37-111.11 ng·m L~(-1). The regression equation of standard curve was y =-1.252 6 x+2.853 2 with the correlation coefficient( R~2) 0.986 6. The total coefficient of variation was 6.97%,and the blank pork recovery rate was 96.90%-118.79%. [Conclusions]The established indirect competitive ELISA method is specific,accurate,precise and reproducible,and can be used for rapid detection of CPZ residues in pork tissues.
