环境雌激素硼对大鼠胸腺组织结构的影响

摘  要:
选用192只28±2d实验用清洁级SD大鼠,分为6组,每组32只,雌雄各半,分别饮用添加硼0、40、80、160、320和640mg/L的蒸馏水,试验期60d,分别于试验第30d(65日龄)和60d(95日龄)每组随机取鼠16只,雌雄各8只,10%水合氯醛腹腔麻醉,立即解剖取胸腺,称重,制作石蜡切片,HE染色,光镜观察并显微摄影。结果显示:与对照组相比较,65日龄试验Ⅲ~Ⅴ组雄鼠和试验Ⅴ组雌鼠胸腺质量显著或极显著降低(P<0.05或P<0.01);95日龄试验Ⅰ组雄鼠胸腺质量显著升高(P<0.05),试验Ⅴ组雄鼠和雌鼠显著或极显著降低(P<0.05或P<0.01)。65日龄和95日龄试验Ⅴ组雄鼠和雌鼠胸腺器官指数显著低于对照组(P<0.05);试验Ⅰ、Ⅱ组胸腺结构表现较明显促进作用;试验Ⅲ、Ⅳ组胸腺结构呈现一定的退化迹象;试验Ⅴ组胸腺结构退化现象明显,实质内淋巴细胞数量明显减少。试验结果表明,饮水添加40、80mg/L硼对大鼠胸腺发育及其组织结构有较明显促进和改善作用;添加160、320mg/L硼对胸腺发育有一定不良影响;添加640mg/L硼对大鼠胸腺发育则有明显的抑制甚至毒性作用。

摘  要:
192 SD rats(28±2-day-old) with no bacteria infection were randomly divided into six groups(n=32,16 male rats and 16female rats).Treated rats drank the water which was supplemented with 40,80,160,320 and 640 mg/L boron,respectively,and the control rats received distilled water for 60 days,respectively.At the 30th and the 60th day of experiment period,16 rats(n=8,male=female) from each group were selected and made into narcosis with 10% Chloral Hydrate.Then the thymuses were obtained and weighed after dissection;then the samples were made into paraffin sections,observed and photographed under Olympus CH-30 microphotograph system.Results are follows: Compared with control group of the same age,the thymus mass of male rats in experiment group from Ⅲ to Ⅴ and female rats in experiment group Ⅴ,and the organs index of thymus of male and female rats in experiment group Ⅴ at 65 day-old decreased significantly or most significantly(P<0.05 or P<0.01);the mass of thymus of male rats in experiment group Ⅰ at 95 day-old increased significantly(P<0.05),but the mass and the organs index of thymus of male and female rats in experiment group Ⅴ decreased significantly(P<0.05).Under the light microscope,the structure of thymus of experiment group Ⅰ and Ⅱ at 65 and 95 day-old has developed well,but the microstructure of thymus of experiment group Ⅲ and Ⅳ exhibited a certain degeneration,and the degeneration of thymus in experiment group Ⅴ was more obvious for lymphocytes decreased markedly.The results indicated that the distilled water supplemented with from 40 to 80 mg/L of boron could promote the thymus development and improve the structure of thymus obviously,but slightly harmful effects,obvious inhibition,or even toxic effect on the development and microstructure of thymus could be observed when the boron supplementation from 160 to 640 mg/L.
