苦瓜枯萎病生防木霉的筛选鉴定及其定殖的qPCR检测

摘  要:
为绿色防控苦瓜枯萎病,本研究从苦瓜根际土中分离出多株木霉,通过平板对峙培养法和盆栽防病试验进行拮抗苦瓜枯萎病菌的木霉菌株筛选,对筛选到的菌株进行形态学及分子生物学鉴定,并运用实时荧光定量PCR技术对其在苦瓜植株及土壤中的定殖能力进行检测.结果表明:筛选得到两株防效较好的木霉菌株M2和MX,对苦瓜枯萎病的防治效果分别达54.63%和45.72%.结合形态学和分子生物学鉴定,确定M2菌株为绿木霉(Trichoderma virens),MX菌株为棘孢木霉(Trichoderma asperellum).木霉菌株M2和MX均不能在苦瓜植株中定殖,但在土壤中可以稳定定殖.两个木霉菌株具有应用于苦瓜枯萎病的田间生物防治潜力.

摘  要:
In order to prevent and control the wilt of bitter gourd caused by Fusarium oxysporum,several Trichoderma strains was isolated from rhizosphere soils of bitter gourd field in this study.Trichoderma strains that had good control effects on the wilt of bitter gourd were screened by plate confrontation test and pot culture test,then the strains were conducted morphological and molecular biological identification.The colonization ability of screened strains of Trichoderma in the plants and soils of bitter gourd was detected by real-time PCR.The results showed that two Trichoderma strains M2 and MX had better control effect on wilt of bitter gourd with the control efficiency of 54.63% and 45.72%,respectively.According to the morphological and molecular biological identification,strain M2 was identified as Trichoderma virens,and strain MX was identified as Trichoderma asperellum.Strain M2 and MX could not be colonized in plants of bitter gourd,but they could be steadily colonized in soils.They could be applied in biocontrol of wilt of bitter gourd in field.
