慈竹组织培养消毒方法筛选及丛芽诱导研究

摘  要:
选择慈竹当年生生长健壮半木质化尚未萌芽的枝条作为外植体,对慈竹组织培养中的消毒方法进行筛选和激素处理及培养基对其丛芽诱导的影响实验。结果表明,将外植体首先用自来水冲洗2 h,然后用70%酒精擦拭1遍,最后用0.1%的升汞消毒2次,每次12 min消毒效果最好,污染率较低,褐化率也较低。不同浓度激素组合下6-BA浓度为3.0 mg/L时,丛芽诱导率最高,达73.11±3.42%。MS培养基丛芽诱导率显著高于3/4 MS培养基和1/2 MS培养基。

摘  要:
Using the semi-lignified branches of Neosinocalamus affinis as the explants,the optimization of disinfection method and multiple shoot induction were studied.The results showed that washing the explants by tap water for 2 hours,scrubbed with 70 % alcohol and then disinfected by 0.1 % mercuric chloride for 12 minute twice could reach good effect,with low contamination rate and browning rate.Among the tested different combination of hormones,treating the explants with 6-BA at the concentration of 3.0 mg/L got the most promising multiple shoot induction,which could reach 73.11±3.42 %.The induction effect of MS culture medium was significantly higher than those of 1/2 MS and 3/4 MS.
