四川干旱干热河谷核桃资源遗传多样性分析

摘  要:
利用AFLP-银染技术,对四川干旱半干旱河谷流域的4个核桃居群和干热河谷流域的2个铁核桃居群共46个样品进行遗传多样性与亲缘关系分析。选择EcoRⅠ/MseⅠ酶切组合,选用多态性高、分辨力强的引物组合E32/M48、E32/M61、E33/M61、E33/M62、E35/M74、E41/M48分别对供试材料的基因组DNA进行选择性扩增,共扩增出475个遗传位点,其中332个多态位点,多态率为69.89%;核桃群体所检出的位点平均有效等位基因数Ae、Nei’s基因多样度H、平均Shannon信息指数I分别为1.2972、0.1745、0.2629,而铁核桃群体分别为1.2758、0.1616、0.2422;从种级水平来看,2物种内部居群间的遗传分化系数Gst分别为0.3794,0.2252,表明核桃群体与铁核桃群体的变异主要存在于居群内,分子方差分析(AMOVA)的结果与以上分析结果一致;主成分分析(PCA)结果与聚类分析的结果相符;从Nei’s遗传距离和遗传一致度分析了各供试资源间的亲缘关系;经SPSS分析表明居群间的遗传距离与地理距离之间存在极显著相关性(R2=0.847**)。从研究结果可得出以下结论,物种的繁育系统、居群间的基因流、地理隔离是该区域核桃及铁核桃居群遗传结构形成的主要原因。

摘  要:
Four populations of Juglans regia L. distributed in arid or semi-arid valleys, Minjiang River and Daduhe River namely, and 2 populations of J. sigillata Dode at Jinshajiang dry-hot valley, a total of 46 samples were selected and analyzed by AFLP-silver staining using 6 pairs of AFLP primers screened, 475 bands including 332 polymorphic bands were ob- tained, the percentage of polymorphic band (P) was 69.89%. As for J. sigillata populations at the species level, the effective number of alleles per locus (Ae) was 1.275 8, and the Nei’s gene diversity index (H) was 0.161 6, Shannon’s information index (I) was 0.242 2. For J. regia populations at species level, the estimates were Ae=1.297 2, H=0.174 5, I=0.262 9. Ge- netic differentiation coefficients among J. regia populations and J. sigillata populations (Gst) were 0.379 4,0.225 2 respec- tively, which showed small partial genetic diversity present among populations of both in two species. This result was ap- proved by AMOVA. Cluster analysis, the dendrogram analysis and the principle component analysis (PCA) for all of the samples and populations were performed. Results obtained through cluster analysis were consistent with PCA results. The UPGMA cluster based on Nei’s distance matrix was analyzed and some further analysis about the relationships among the populations was made. There was a significant association between genetic and geographical distances (Pearson coefficient was 0.847**) among Juglans populations analyzed by SPSS software. The possible reasons for the genetic diversity and ge- netic structure of the six populations in this region were mainly related to the geographic isolation, inbreeding and gene flow of the populations.
