近期泰安周边地区鸭病毒性肝炎流行情况调查

摘  要:
为探究泰安周边地区接种鸭病毒性肝炎疫苗或打了卵黄抗体的鸭群仍然发生病毒性肝炎的真正原因,我们对2014年4~8月份送检的10份疑似鸭病毒性肝炎病例进行了剖检诊断、组织病理学观察及RT-PCR检测,并进行遗传进化及同源性分析。结果显示,10份病例在临床症状及病理学变化上均符合鸭病毒性肝炎的特征,但只有7份病例PCR诊断为DHV阳性,3份为阴性。这7份病毒的VPO基因同源性在98.6%~100%,与DHV-A、DHV-B处于不同分支,同源性均不足76%,与中国分离株JS2010、C-YCW处于较小的分支上,同源性在98.3%~98.9%,亲缘关系最近,但为不同小分支,表明我们检测到的鸭病毒性肝炎毒株为DHV-C型,但与中国分离株JS2010、C-YCW存在一定差异性。以上结果表明,DHV-C已成为鲁中地区当前流行的优势毒株,注射针对DHV-A的疫苗或卵黄抗体无效现象的发生可能与此有关。

摘  要:
In the present study we investigated the prevalence and molecular characterization of Duck hepatitis virus in and around Tai'an area. From the suspected duck population we collected about 10 samples during the period of April to August2014. Initially these samples were evaluated by necrscopsy, histopathological observation and RT-PCR findings. For further confirmatory studies these samples were subjected to genetic evaluation and homology analysis. The clinical symptoms and histopathological changes revealed that all the samples showed the presence of duck virus hepatitis. Out of the total samples seven were conformed for the presence of viral RNA by RT-PCR. The genetic evaluation and homology analysis of the positive sample on VP0 gene was 98.6%~100%, showed close similarities with previously reported Chinese isolates JS2010 and C-YCW homologies, 98.3%-98.9%. Although these isolates having close phylogenetic relationship but still having some variation in the small branches, which suggested that the present isolates having certain difference from the previous isolates.It is clear that DHV isolate from all the positive samples having less than 76% similarity with DHV-A and DHV-B. Taken together, results suggest that DHV isolated from Shandong province of china is different from the early isolated DHV-A and DHV-B type and is conceder as a new DHV type(DHV-C). Thus for the better control of this economically important disease in Shandong province, it is necessary to use the newly isolated strain for Vaccine preparation.
