利用人工合成小麦自发产生的非整倍体进行基因定位

摘  要:
人工合成小麦可以同时将四倍体小麦和节节麦的优良基因转移到六倍体遗传背景。人工合成小麦可通过两种途径自发产生非整倍体:一是通过减数分裂核再组形成的非整倍配子结合,在合成小麦的第一代就产生非整倍体,另一种是通过新形成人工合成小麦的细胞学不稳定,产生非整倍体。人工合成小麦自发产生的非整倍体,可以专门用于四倍体小麦或节节麦质量性状基因的染色体定位。该细胞学定位方法主要包括以下几个步骤:①筛选出具有目标性状基因的四倍体小麦或节节麦材料;②创制人工合成小麦;③筛选目标性状存在差异的植株;④鉴定缺失染色体。该方法对四倍体小麦或节节麦目标基因转移的同时,实现对目标基因的染色体定位。

摘  要:
Genes in Triticum turgidum and Aegilops tauschii can be simultaneously transferred to Synthetic hexaploid wheat(SHW).SHW aneuploids may be spontaneously formed by the union of aneuhaploid gametes,derived from meiotic restitution from the F1 hybrids of T.turgidum with Ae.tauschii,or produced by cytological instability of new SHWs.These SHW aneuploids are useful in chromosomal location of genes specific for T.turgidum and Ae.tauschii.This cytological method for gene location involves four main steps: ①screening out T.turgidum or Ae.tauschii genotype with desirable gene;②producing new SHWs by hybrids of T.turgidum with Ae.tauschii;③finding the SHW plants with contrast phenotypes for analyzed character;④identifying SHW aneuploids lacking the chromosome carrying desirable gene.This method can conduct two tasks,transferring gene from T.turgidum or Ae.tauschii to hexaploid wheat and locating the transferred gene.
