哲罗鲑(Hucho taimen)卵巢发育过程中下丘脑-垂体-性腺轴(HPG轴)相关因子的动态变化

摘  要:
采用酶联免疫吸附法(ELISA)测定卵巢处于不同发育期的7龄雌哲罗鲑Hucho taimen血清中雌二醇(E_2)与睾酮(T)的含量,检测下丘脑促性腺释放素(GnRH)、垂体促滤泡激素(FSH)与促黄体激素(LH)及肝脏卵黄蛋白原(Vtg)mRNA表达水平的变化,分析生殖激素基因、性腺指数(GSI)和性类固醇激素之间的相关性,以深入了解下丘脑-垂体-性腺轴(HPG轴)对哲罗鲑卵巢发育的调控作用及其机制。结果显示,在卵巢发育期内下丘脑GnRH mRNA表达水平在Ⅳ早期时显著升高,至Ⅳ晚期时达到最低,随后显著升高;垂体FSH与LH mRNA表达水平自Ⅲ期显著降低,随后显著升高,至Ⅳ晚期达到峰值后逐渐下降;肝脏Vtg mRNA表达水平自Ⅳ早期显著升高,在Ⅳ期中达峰值,随后下降至Ⅱ期、Ⅲ期水平。相关性分析表明,下丘脑GnRH mRNA表达水平与GSI呈正相关;与FSH、LH mRNA的表达水平呈负相关,而垂体FSH与LH mRNA之间则呈现显著正相关(P<0.05)。血清中E_2与T含量变化趋势相似,均与卵母细胞同步发展。GnRH、FSH与LH变化的规律性确保了HPG轴对Vtg、性类固醇激素(E_2与T)等合成与分泌的调控。本研究可为系统、科学地开展哲罗鲑人工繁育与健康养殖提供理论依据。

摘  要:
In order to understand the mechanisms of hypothalamus-pituitary-gonadal axis(HPG axis)for reproduction regulation, the expression levels of gonadotropin-releasing hormone(GnRH)gene of hypothalamus, follicle stimulating hormone(FSH)gene and luteinizing hormone(LH)gene of pituitary, and vitellogenin(Vtg)gene of liver, as well as the contents of serum estradiol(E_2)and testosterone(T), were determined in 7 years old female taimen(Hucho taimen)during the different ovarian developmental stages. The correlations among the reproductive hormone genes, gonado-somatic index(GSI)and sex steroid hormones were analyzed. The expression levels of GnRH gene significantly increased at early stage Ⅳ reached to the lowest at late stage Ⅳ and then significantly increased during the ovarian development. The expression levels of FSH and LH genes significantly declined at stage Ⅲ, reached to the peak value at late stage Ⅳ and then gradually decreased during the ovarian development(P < 0.05). The expression levels of Vtg in liver were significantly increased from early stage Ⅳ, reached to the peak value at mid-stage Ⅳ and then decreased to the levels of stages Ⅱ and Ⅲ.Correlation analysis revealed that the expression levels of GnRH were positively related with GSI and negatively with FSH and LH expression levels. There was positive relationship with the expression levels between FSH and LH(P < 0.05). The changes of E_2 and T contents in serum were synchronous with the oocyte development. The results indicated that the regular changes in expression levels of Gn RH, FSH and LH ensured the roles of HPG axis in modulating the synthesis and secretion of Vtg and sex steroid hormones(E_2 and T), which would be helpful to systematically and scientifically carry out artificial and healthy breeding of taimen.
