白头翁汤中总多糖对微血管内皮细胞糖萼糖链表达的影响

摘  要:
【目的】研究白头翁汤中总多糖对微血管内皮细胞(microvascular endothelial cells,MVECs)糖萼糖链的影响。【方法】体外培养猪小肠黏膜MVECs,以50μg/m L白头翁汤中总多糖刺激48 h后,利用凝集素荧光技术检测刀豆凝集素(concanavalin A,Con A)、麦胚凝集素(wheat germ agglutinin,WGA)、双花扁豆凝集素(dolichos bifows agglutinin,DBA)、荆豆凝集素(ulex europaeus agglutinin I,UEA I)、花生凝集素(peanut agglutinin,PNA)、雪花莲凝集素(galanthus nivalis lectin,GNL)、番茄凝集素(lycopersicon esculentum lectin,LEL)受体糖链的表达。【结果】正常情况下猪小肠黏膜MVECs WGA、LEL、Con A和GNL四种凝集素荧光染色呈强阳性,DBA和PNA荧光染色呈弱阳性,UEA I荧光染色呈阴性;白头翁汤中总多糖能显著提高WGA和LEL荧光染色的强度。【结论】白头翁汤中总多糖能上调猪小肠黏膜MVECs表达N-乙酰氨基葡萄糖。

摘  要:
【Objective】To investigate effects of polysaccharides from pulsatilla decoction on sugar chain expression in microvascular endothelial glycocalyx. 【Methods】Porcine intestinal mucosa microvascular endothelial cells( MVECs) in vitro were cultured in vitro,and were treated with 50 μg / m L polysaccharides from pulsatilla decoction for 48 hour. Then,the receptor sugar chains of concanavalin A( Con A),wheat germ agglutinin( WGA),dolichos bifows agglutinin( DBA),ulex europaeus agglutinin I( UEA I),peanut agglutinin( PNA),galanthus nivalis lectin( GNL),lycopersicon esculentum lectin( LEL) in MVECs were detected by lectin fluorescent staining. 【Results】Normal porcine intestinal mucosa MVECs displayed intensive fluorescence for WGA,LEL,Con A and GNL staining,weak fluorescence for PNA and DBA,and negative for UEA I staining. Treatment with polysaccharides from pulsatilla decoction enhanced the fluorescence intensity of WGA and LEL staining. 【Conclusion】Expression of N-Acetylglucosamine in porcine intestinal mucosa MVECs could be increased by polysaccharides from pulsatilla decoction.
