黄连素抑制猪流行性腹泻病毒复制和组装

摘  要:
通过Western blot、qRT-PCR、噬斑形成试验等方法检测细胞病变、病毒增殖水平、细胞中病毒蛋白水平与基因水平表达来探究黄连素是否具有抗猪流行性腹泻病毒(porcine epidemic diarrhea virus,PEDV)活性,并通过检测细胞凋亡相关的标志蛋白PARP1、caspase7以探究黄连素是否影响病毒引起的细胞凋亡。结果显示,黄连素明显抑制了PEDV在Vero细胞中引起的细胞病变,明显降低了细胞内与上清中的病毒粒子的产生。在后续研究中,发现黄连素抑制了PEDV的复制和组装阶段,对病毒的吸附、入胞、释放的过程并没有明显影响。另外,检测了细胞凋亡的标志蛋白PARP1和caspase7,发现黄连素下调了PARP1、caspase7的剪切,减弱了PEDV诱导的细胞凋亡。结果表明,黄连素具有抗PEDV活性,并抑制PEDV的复制与组装阶段,可作为一种针对PEDV感染的潜在抗病毒药物。

摘  要:
Porcine epidemic diarrhea virus(PEDV) can infects newborn piglets and causes huge economic losses to the pig industry.However,no effective PEDV vaccine available.It is of great significance to develop antiviral drugs.Berberine(C20 H19 NO4.Cl),a natural isoquinoline alkaloid extracted from traditional Chinese medicine,shows antiviral activity during various viral infections.In order to explore whether the Chinese medicine berberine has antiviral effect against porcine epidemic diarrhea(PED).Western blot,qRT-PCR,plaque formation assay were used to detect cytopathic effect,virus proliferation,viral protein level and gene expression level in Vero cells to explore whether Berberine has anti-PEDV activity and detect apoptosis-related marker proteins,PARP1,caspase7 to explore whether berberine affects apoptosis caused by the PEDV.The results showed that berberine significantly inhibited the cytopathic effect induced by PEDV,and reduced the production of virions in the cells and supernatants.In the follow-up study,it was found that berberine inhibited the replication and assembly phase of PEDV,and had no significant effect on the process of virus adsorption,entry and release.In addition,the marker proteins of apoptosis,PARP1 and caspase7 were detected,and it was found that berberine attenuated PEDV-induced apoptosis.It was first discovered that berberine has anti-PEDV activity and inhibits PEDV replication and assembly,suggesting that berberine may be a potential antiviral drug against PEDV.
