米胚芽中α-生育酚的高效液相色谱法测定

摘要：

应用反相高效液相色谱法测定了米胚中α-生育酚的含量。样品用无水乙醇索氏法提取后直接加固体氢氧化钾皂化。皂化物经石油醚液液分配、浓缩和转溶于乙醇后,用具C_(18)分离柱—甲醇流动相—292nmUV检测的色谱系统分析,测得一般粗米糠胚中α-生育酚含量为19.94±0.49mg/100g,方法回收率为95±4％,浓度范围在65ng～320ng之间呈线性。

摘要：

This paper describes the application of high performance liquid chromato-graphy(HPLC)to the determination of cc-tocopherol in rice germ.The samples were pretreated by the following main steps-the soxhlet extraction with absolute alcohol,saponification.Liquid-Iiuqid distribution with petroleum ether,evaporation and redissolve in alcohol.The alcoholic extracts were analysed by HPLC with C18 column,a mobile phase of 100% methanol and the UV detection of 292nm.The recovery and content of a-tocopherol obtained by using this method are 95±4% and 15.94±0.49mg/100g,respectively.
