匍匐小菊遗传转化过程中选择压力的确定

该试验以北京农科院生物中心新近培育的匍匐小菊品种"粉地毯"为材料,以叶片为外植体,建立"粉匍"的高效再生体系。试验共进行了6种不同培养基的筛选,研究了6-BA对于再生频率的影响,最终确立了6-BA的最佳浓度。在MS+6-BA(0.5 mg/L)+NAA(0.1 mg/L)培养基上获得91%的再生率,在1/2 MS+NAA(0.1 mg/L)培养基上诱导生根。在获得高效叶盘再生体系的基础上,又研究了植物遗传转化过程中常用的2种选择标记卡那霉素(Km)和除草剂(PPT)对菊花不定芽分化的影响。结果表明:大于8 mg/L的Km和大于0.4 mg/L的PPT能够完全抑制试验材料的不定芽分化。试验最终确定了匍匐小菊对卡那霉素、草丁膦的适宜筛选浓度,分别为5 mg/L和0.2 mg/L,为以后进行匍匐小菊的基因工程工作奠定了基础。

摘  要:
High regeneration efficiency system of sprawling Dendranthema morifolium new cultivar"pink carpet" bred recently by Beijing Agro-Biotechnology Research Center was set up with leaf discs as explants.Six different medium were tried to investigate the effects of 6-BA on the Dendranthema regeneration frequency.Proper concentration of 6-BA was determined.Shoots regenernated from 91% of the total explants in the medium MS+6-BA(0.5 mg/L)+NAA(0.1 mg/L).Roots were induced in the medium 1/2 MS+ NAA(0.1 mg/L).After the acquirement of high regeneration frequency system with leaf discs,effects of two commonly used selecting agents in plant genetic transformation,kanamycin(Km)and phosphinothricin(PPT),on the differentiation of adventitious shoots from in vitro leaf discs of Dendranthema morifolium were investigated.The results indicated both Km and PPT completely inhibited shoot induction when Km concentration was higher than 8 mg/L or PPT concentration was higher than 0.4 mg/L.In this paper proper concentration of Km and PPT for selection pressure of genetic transformation was determined as 5 mg/L and 0.2 mg/L individually.This research established a foundation for genetic engineering of sprawling Dendranthema morifolium.
