广东湛江杂草稻qSH1基因片段序列分析

摘  要:
为了探明杂草稻与栽培稻qSH1基因片段的序列是否存在差异,为研究杂草稻的落粒性提供依据。利用已经报道的扩增水稻qSH1基因的引物,对来源于广东湛江雷州地区的6份落粒性极强的杂草稻进行PCR检测,通过测序得到长度约460bp的核苷酸碱基序列,将获得的序列进行比对分析,序列分析结果表明:扩增所获片段大致位于qSH1基因下游100kb处,其碱基序列与NCBI网站公布的水稻该片段序列为同源序列,同源性达100%。这6份杂草稻与水稻之间共同存在一个碱基位点差异,经证实为前人报道的SNP位点。另外6份杂草稻所获片段序列不完全一致,存在一个位点碱基差异,但不能确定是否为一个作用位点。

摘  要:
The purpose was to explore whether differences exist in qSH1 gene between weedy rice and Oryza sativa , and provide a basis for studying the seed shattering of weedy rice. Using the primers that had been used to amplify the qSH1 gene of Oryza sativa, PCR was done with 6 samples of weedy rice which have strong seed shattering habit and origin from Zhanjiang of Guangdong province, about 460 bp of nucleotide bases fragments were obtained after sequenced, the result of sequences alignment showed that the fragment located at region approximately 100 kb downstream of qSH1, and was homologous with what has collected on the NCBI website, the homology value was up to 100% . There was a nucleotide site difference between the 6 weedy rices and Oryza sativa, which was proved to be the SNP site reported before. At the same time, not all the nucleotide sequences that derived from the 6 weedy rices are the same, there is a different nucleotide site, but can ' t determine whether it is an action site.
