水提山茱萸多糖的理化性质及抗氧化活性研究

摘  要:
山茱萸经热水抽提、Sevag法脱蛋白、乙醇沉淀、DEAE-纤维素柱层析分离,再通过Sephadex G-200柱层析进一步纯化,得到白色粉末状多糖PFCAIII。对该多糖进行完全酸水解、薄层层析、红外光谱分析,结果证明此多糖由鼠李糖、阿拉伯糖和葡萄糖以13.74:50.54:35.72的摩尔比组成,平均分子量为1.74×104。该多糖可以有效抑制猪油和芝麻油的氧化,并对邻苯三酚自氧化系统产生的超氧阴离子自由基(O2-·)具有一定的清除能力。用Fenton反应测定PFCAIII对羟基自由基(·OH)的清除作用,对·OH的清除率达50%所需浓度(EC50)为430μg/ml。

摘  要:
The polysaccharide PFCA III was isolated from Fructus Corni through water extraction,de-protein ethanolprecipitating and DEAE-cellulose column chromatographic fractionating.It was further purified by Sephadex G-200 columnchromatography.The molecular weight of PFCA III was 1.74×104.Chemical composition analysis by TLC and HPLC showedthat it was composed of rhamnose, arabinose and glucose in a molar ratio of 13.74:50.54:35.72 respectively.Thisheteropolysaccharide PFCA III had the activity of oxidation resistance to axunge and gingili.It also had an action to scavenginghydroxyl free radicals (·OH) generated by Fenton reaction and superoxide anion (O2-·) generated by pyrogallol autoxidationsystem.The scavenging activity to hydroxyl free radicals and superoxide anion was represented by the content scavenged 50%of all radicals (EC50)of 430μg/ml.
