家蚕α淀粉酶基因的多态性研究

摘  要:
【目的】从分子水平研究不同家蚕品种间淀粉酶基因多态性差异,为家蚕的育种选择和遗传进化研究提供有力的证据。【方法】根据网上基因数据库中的家蚕α淀粉酶基因序列(序列号:U07847),在第4和第5外显子区设计了一对特异性引物,在1个镇江野蚕和6个不同的家蚕品种基因组内进行PCR扩增,发现了该基因区域在不同品种间的多态现象。【结果】根据所得基因测序结果信息,用Clustalx软件进行分析并构建了分子系统树。其聚类结果在一定程度上也比较符合家蚕地理和化性的分布,而且镇江野蚕和家蚕品种距离较远。【结论】家蚕淀粉酶是一个为家蚕的起源和进化密切相关的分子标记基因,值得更深入的研究和探讨。

摘  要:
【Objective】The study determined genetic relationships among strains of silkworm,Bombyx mori,the polymorphism ofαamylase gene was studied in molecular level.【Method】We designed a pair of sequence specific primers from the silkwormα amylase gene that has been published on Genbank(accession number: U07847).After amplification in six silkworm strains and one Chinese mandarina,polymorphism was been detected.【Result】 Furthermore,the genetic diversity and phylogenetic relationship of the seven strains was investigated using the Clustalx software,which was based on the gene sequences information.The analysis grouped these strains roughly according to their voltinism and geographical origin characters.The Chinese silkworm strains seemed to be differentiated in the earliest year from Chinese the mandarina strain.【Conclusion】The silkworm amylase gene was proved to be a strong marker in the genetic diversity study,and it required further investigation and discussion.
