果梅(Prunus mume)品种的荧光AFLP指纹分析

摘  要:
荧光AFLP以其操作简易、灵敏性高、可半自动化操作、可软件辅助分析等特点在果树品种鉴定以及遗传资源分析中具有明显的优越性。对荧光AFLP用于果梅品种鉴定进行了研究。所利用的8对选择性引物都可以获得清晰的AFLP指纹,并且任何一对引物都能够很好的区分这50个品种,表明了该技术适于快速鉴定大量果梅品种或用于种质资源的遗传分析。同时,研究利用AFLP指纹信息对来源我国江苏、浙江、广东、福建、云南、四川、湖南7个省份以及日本的果梅品种的遗传多样性指数进行了分析,结果表明,日本(0.2451),中国江苏、浙江(0.1912)果梅的遗传多样性水平明显高于其他几个省份,可能由于这2个地区果梅品种的来源渠道多或者存在其他物种的基因渗入。

摘  要:
Florescent AFLP,which has the characteristics of easy operation,high sensitivity,semi-automatic,and gel-reading with software,can show obvious advantages in genotyping accessions of fruit crops. 8 primer pairs were used to genotype 50 mei cultivars in this study,and all these primers could generate clear fingerprints,based on which each primer pair could be used to separate these 50 cultivars well. The Genetic Diversity Index (GDI) of the cultivars from different provinces showed that the mei in Japan, and China (Jiangsu, Zhejiang) had higher genetic variation, of which their GDI were 0.245 1 and 0.1912. This might be due to the various origins and there might have happened some gene flow between mei and some other plant species.
