中度嗜盐菌ST77及其萃取物对马铃薯干腐病的防效及菌株鉴定

摘  要:
采用平板对峙法和牛津杯法测定菌株ST77及其3种有机溶剂(氯仿、乙酸乙酯、正丁醇)萃取物对马铃薯干腐病的抑菌活性;通过活体试验,测定菌株ST77发酵液及其3种有机溶剂萃取物对马铃薯干腐病的活体防效,并进行安全性评价;采用形态学观察、生理生化特征鉴定及分子生物学方法对菌株ST77进行鉴定.结果表明:中度嗜盐菌ST77及其萃取物对马铃薯干腐病病原真菌青9A-4-13、青9A-5-2和65B-2-6具有不同程度的抑制作用,抑菌圈直径最大为0.60 cm;中度嗜盐菌ST77发酵液及其萃取物能够完全抑制马铃薯活体上病原真菌65B-2-6的生长,坏果率为零,对其他病原真菌具有不同程度的抑制作用.其中,中度嗜盐菌ST77正丁醇萃取物对马铃薯干腐病病原真菌的抑制活性较高;同时,ST77发酵液及其正丁醇萃取物对马铃薯储藏安全.经鉴定,菌株ST77为短小芽孢杆菌(Bacillus pumilus).

摘  要:
The anti-fungi activities of strain ST77 and its extracts from 3 organic solvents(chloroform, ethyl acetate, n-butanol) were determined by confrontation culture and Oxford cup method. The preventive effects of strain ST77 fermented broth and its extracts from the 3 organic solvents against potato dry rot were determined in vivo and followed by safety test. Strain ST77 was further characterized regarding morphological, physiological and biochemistry, and molecular biology features. The results showed that the moderately halophilic strain ST77 and its extracts had different inhibitory activities against potato dry rot pathogens of Qing 9 A-4-13, Qing 9 A-5-2, and 65 B-2-6, with the maximum inhibition zone diameter being 0.60 cm. In vitro pathogenicity test on potato demonstrated that pathogen 65 B-2-6 was completely inhibited without bad fruit by strain ST77 fermented broth and its extracts, and to a lesser extent the other 2 pathogens were also inhibited in varying degrees. Among them, the extract of ST77 by n-butanol had higher inhibitory activity against Fusarium spp., while ST77 fermented broth and its n-butanol extract were safe for potato storage. Lastly, strain ST77 was identified as Bacillus pumilus.
