猪供核细胞不同处理对细胞凋亡及重构胚体外发育的影响

摘  要:
在核移植过程中,为了选择出一种最优的供核细胞处理方法,采用接触抑制、血清饥饿以及75 nmol/L曲古抑菌素A(Trichostatin A,TSA)3种方法分别对猪体细胞进行处理。通过AnnexinV-FITC法检测3种处理对细胞凋亡的影响,并将3种方法分别处理过的体细胞作为供核细胞,利用体细胞核移植技术(Somatic cell nuclear transfer,SCNT)研究供核细胞不同处理方法对猪重构胚体外发育能力的影响。结果表明:接触抑制和75 nmol/L TSA处理组对细胞的早期凋亡、晚期凋亡以及细胞坏死率同对照组比较差异不显著,而血清饥饿处理组无论是早期凋亡、晚期凋亡还是细胞坏死率均显著高于对照组。3种方法处理供核细胞,接触抑制处理组卵裂率同血清饥饿处理组比较,差异不显著,但是其显著高于75 nmol/L TSA处理组。3种方法处理的重构胚囊胚率差异不显著。接触抑制方法处理供核细胞相对于其他2个处理组更有利于重构胚的体外发育。

摘  要:
In order to choose the best way to pretreat the donor cells in the process of SCNT,porcine somatic cells were treated with contact inhibition,serum starvation and 75 nmol /L TSA,respectively. By AnnexinV- FITC method,the effects of three methods on cell apoptosis were detected and by somatic cells nuclear transfer technique,the effects of different treatments of donor cells on reconstructed embryo development were studied. The developmental competence of SCNT embryos was assessed by rate of cleavage and blastocyst formation. The results indicated that the percentage of early,advanced and necrosis in serum starvation treatment was significantly higher than that of any other treatment. The rate of cleavage and blastocyst formation in contact inhibition treatment was higher than that of any other treatment. Above all,the results suggested that contact inhibition treatment had lower percentage of apoptosis and higher developmental competence of SCNT embryos.
