利用16S rDNA序列分析鉴定一株产抗菌物质的微生物菌株

摘  要:
从桃子上分离纯化得到1株产抗菌物质的菌株,命名为fmb3菌,通过抑菌实验发现该菌对革兰氏阳性菌蜡样芽孢杆菌、革兰氏阴性菌大肠埃希菌和真菌扩展青霉等3种常见食品病原菌具有显著的抑菌效果。利用16SrDNA分析对该菌作了系统进化鉴定,首先提取fmb3菌基因组DNA,根据不同种属的细菌的16SrDNA序列两端的保守性设计通用引物,对fmb3菌16SrDNA片断进行PCR扩增,对扩增得到的目标片段测序,并且利用NCBIBLAST对测序结果进行比对,构建系统进化树进行分析,初步确定该菌属于枯草芽孢杆菌。

摘  要:
A microorganism strain producing the antimicrobial substance was isolated from peaches and was named fmb3. The inhibitory test showed that the antimicrobial substance produced by fmb3 can strongly inhibit the gram positive strains Staphy lococcus aureus and Bacillus cereus,the gram negative strain Escherichia coli and the mold Penicillium expansum. Identification of the microorganism strain was also performed using the 16S rDNA sequence analysis. First the genomic DNA from fmb3 was extracted,and then the general primers were designed according to conservative sequence of bacteria 16S rDNA. Furthermore,amplifying the target 16S rDNA sequence and the sequence analysis by using the NCBI BLAST database was described. The strain was finally identified as the Bacillus subtilis.
