葵花粕中11S球蛋白的分离制备及性质

摘  要:
本文以水酶法提取葵花籽油后的葵花粕为原料,通过盐溶盐析法分离其主要蛋白成分11S球蛋白,并对葵花11S球蛋白进行凝胶过滤纯化与分析、组分分析、氨基酸成分分析和电泳分析。结果显示葵花11S球蛋白有一个单一的峰,其分子量约为380×103;其氨基酸组成与FAO必须氨基酸需要量模式相比赖氨酸含量较低,而其它氨基酸较丰富;葵花11S球蛋白由9条主带和一条微带组成。差示扫描量热仪(DSC)分析表明葵花11S球蛋白干粉和稀溶液的变性温度分别为123.5℃和95.9℃;其荧光发射光谱最大荧光强度在345nm左右;11S球蛋白的二级结构为β-折叠占82.2%,无规则卷曲占17.8%。

摘  要:
11S globular proteins from sunflower meal after defatted by the method of aqueous enzymatic extraction were obtained and analyzed for gel filtration chromatography,composition,amino acid content and gel electrophoresis spectrum in this paper.The results showed that 11S Globular proteins contained only one fraction having molecular weights of 380×10 3 ,Compared with FAO reference pattern,it possesses adequate amounts of essential amino acids except the slightly inadequate lysine,11S sunflower globular proteins have 9 major bands and one minor band.Differential Scanning Calorimetry(DSC)analysis showed that the denaturation temperatures of powder and solution of 11S sunflower globular proteins were 123.5℃and 95.9℃,respectively.The fluorescence emission spectrum of 11S sunflower globular proteins gave a maximum at 345nm.The secondary structure analyed by circular dichroism(CD)revealed thatβ-sheet and random coil were 82.2%and 17.8%,respectively.
