某河南斗鸡原种场禽白血病病毒感染状况监测

摘  要：
为了解某河南斗鸡原种场禽白血病病毒(ALV)感染状况,分别于2017年1、4、9、12月和2018年7月随机或全群采集该场核心鸡群雏鸡胎粪棉拭子40份、泄殖腔棉拭子973份、血清150份以及留种用公鸡精液42份,采用ELISA方法分别检测雏鸡胎粪棉拭子、泄殖腔棉拭子、精液的ALV-p27抗原和血清样品的ALV-A/B、ALV-J亚型抗体.采集33份泄殖腔棉拭子ALV-p27阳性鸡的血浆,接种DF-1细胞,分离外源性ALV,并对分离株env基因进行了序列测定和同源性分析.结果显示,初次检测的雏鸡胎粪棉拭子ALV-p27阳性率为0,之后3次检测的成年鸡泄殖腔棉拭子ALV-p27抗原阳性率分别为84.48%、88%和78.10%;血清ALV-A/B、ALV-J亚型抗体阳性率两次检测结果分别为17.14%、0%和79.31%、24.14%;留种用公鸡精液ALV-p27抗原阳性率9.52%.DF-1细胞培养共分离到6株ALV,其中2株env基因序列与ALV-A亚型SDAU09E2株、ALV-J亚型HPRS103株同源性分别为89.2%、56.4%和89.2%、56.0%.表明该核心鸡群ALV感染率较高,且以A亚型为主.

摘  要：
To get acquainted with the infection situation of avian leukosis virus(ALV)in a Henan game chicken breeds,40 Cloacal swabs s of chicks meconium,973 Cloacal swabs and 150 serum samples were collected randomly or wholly from the nucleus breeds,on month 1,4,9,12 of 2017,respectively.And 42 semen samples of selected breeder cock were also collected on month 7 of 2018.Then ALV-p27 antigen and ALV-A/B or ALV-J antibody were detected by ELISA.33 plasma samples of chickens with Cloacal swab ALV-p27 positive were also collected,and were inoculated on DF-1 cells respectively.Subsequently,env genes of 2 isolated ALV strains were analyzed by MegAlign.The results showed that,the positive rate of ALV-p27 of chicks meconium Cloacal swabs was 0,while the positive rate of ALV-p27 of the adult chicken Cloacal swabs were 84.48%,88%and 78.10%,respectively.The positive rates of ALV-A/B and ALV-J subgroup antibody in serum were 17.14%,0%,79.31%and 24.14%,respectively.The homologous of evn genes between the two strains isolated in this study was 90.5%,and homologous with SDAU09E2 strain of ALV-A subgroup and HPRS103 strain of ALV-J subgroup were 90.1%,56.5%,91.3%and 56.7%,respectively.Together,these data indicated that the nucleus flock of Henan game chickens were infected with ALV highly,and it was mainly infected with ALV subgroup A.
