摘  要:
为研究致鹅卵黄性腹膜炎大肠杆菌(E.coli)fimFGH基因的变异情况,本实验根据GenBank中登录的序列设计引物,PCR扩增了3株致鹅卵黄性腹膜炎性E.coli E0238、E0239、E0240Ⅰ型菌毛的fimFGH基因并测序。序列比对分析结果表明,3株细菌的fimF、fimG和fimH基因及其所编码的蛋白序列同源性达96%以上,其中fimH编码蛋白的序列同源性达98%以上。序列变异分析结果显示,FimH蛋白的变异主要集中分布在FimH粘附素N末端的4个氨基酸位点N91(S)/S91N99、A48V以及N156K,仅有E0239菌株发生了A48V变异;FimF、FimG蛋白各有一个多变区域,分别位于FimF蛋白的111位~124位氨基酸和FimG蛋白的4位~16位氨基酸之间,其中E0238、E0239菌株FimF蛋白发生了A111S的变异,E0238和E0240菌株FimG蛋白发生了C4R/V9L的变异,但变异的具体意义还需要进一步研究。

摘  要:
To analysis the fimFGH genes of Escherichia coli isolated from geese associated with yolk-sac peritonitis,a pair of the specific primers were designed and synthesized according to the fimFGH gene in GenBank.The fimFGH genes in E.coli isolates of E0238,E0239 and E0240 were amplified and sequenced.The sequence analysis demonstrated that fimF,fimG and fimH genes were over 96% homology among the 3 E.coli isolates.Of which the deduced amino acid sequence of fimH gene was up to 98% identity and 4 major mutations were found at the N terminal of FimH,including N91(S) /S91N99,A48V and N156K,but only A48V in E0239 isolate.Basing on the multiple amino acid sequence alignment,there was only one variable region of FimF and FimG respectively,which were located from aa 111 to aa 124 in FimF and aa 4 to aa 16 in FimG.Among the isolates,A111S mutation in FimF was found in both E0238 and E0239 isolates,and C4R/V9L mutation exited in E0238 and E0240 isolates.The biofunctional of these mutations need be further investigated.
