农杆菌介导的Vf基因转化柱型苹果的研究

摘  要:
以柱型苹果“鲁加6号”为试材,研究了根癌农杆菌介导的Vf基因转化条件。研究结果表明,该转化的优化条件为:共培养时间为3 d;延迟培养阶段头孢霉素浓度为200 mg/L,时间为6 d;选择培养阶段,芽分化过程卡那霉素浓度为15 mg/L,生根过程卡那霉素浓度为10 mg/L。PCR检测结果初步表明,外源Vf基因已导入转化植株基因组中。

摘  要:
Transformation systems of columnar apple 'Lujia 6'mediated by A.tumefacience were studied.The result showed that the optimal transformation system was 3 d co-cultivation time,200 mg/L Cef in restoration medium for 6 d,15 mg/L Kan in selection medium during shoot differentiation and 10 mg/L Kan during root formation.PCR analysis result confirmed that Vf gene was integrated successfully into apple genome.
