摘 要:
以酶反应速率受温度影响为理论基础,研究用于监测环境温度的时间-温度指示卡(time-temperature indicator,TTI)。试验选择了适合碱性脂肪酶的反应体系,最终确定了碱性脂肪酶型TTI反应体系中的参数:1 g.L-1碱性脂肪酶0.1 mL,三乙酸甘油酯0.5 mL为反应底物,20 g.L-1聚乙烯醇(polyvinyl alcohol,PVA)9.5 mL作为乳化剂,pH10.6的0.05 mol.L-1Gly-NaOH缓冲液39 mL,1 mol.L-1氯化钙溶液0.7 mL,酚红-酚酞-百里酚酞混合指示剂0.2 mL。通过测定反应体系在不同温度条件下的反应速率,最终确定反应体系的活化能为44.85 kJ.mol-1,该反应体系可以应用于检测以酶促反应、脂肪氧化等为主要引起食品变质因素的食品中。

摘 要:
The time-temperature indicator(TTI)used to monitor the environmental temperature was studied,which is based on the theory that the enzyme reaction rate was influenced by the temperature.Through the experiment,the most optimal reacting system of the alkaline lipase was established.Finally,parameters were developed in the reaction system of alkaline lipase TTI:0.1 mL 1 g·L-1 alkaline lipase,0.5 mL triacetin as enzyme-substrate,9.5 mL 20 g·L-1 polyvinyl alcohol(PVA)as emulsifier,39 mL Gly-NaOH buffer 0.05 mol·L-1 at pH 10.6,0.7 mL 1 mol·L-1 calcium chloride solution,0.2 mL mixture indicator which contained phenol red,phenolphthalein,and thymolphthalein.The activation energy of the TTI reaction system from the different temperature coefficient of the rate constants was found to be 44.85 kJ·mol-1,and this system can be applied to monitor the food quality,which declined because of enzymic and lipid oxidation.
