芦笋不同采收模式下产量、皂苷含量及皂苷合成基因HMGS,FPPS和SS表达研究

摘  要:
为研究不同采收模式对芦笋产量、皂苷量及其皂合成基因表达的影响,通过对弥勒三年生芦笋(泽西骑士),进行白芦笋、光头绿芦笋、留母茎绿芦笋3种芦笋采收方式的小区试验,测定小区产量并收集芦笋样品;测定样品中总皂苷及薯蓣皂苷元的含量;设计引物,RT-PCR克隆皂苷合成中的羟甲基戊二酰辅酶A合酶(Ao HMGS)、发尼烯合酶(Ao FPPS)和沙烯合酶(Ao SS)全长编码序列,并检测这些基因在不同采收模式和不同部位的表达。结果表明:相对于白芦笋,光头采绿芦笋与留母茎采绿芦笋获得较高产量,后者采收芦笋含有最高总皂苷与薯蓣皂苷元。q-RT-PCR分析表明:不同采收模式芦笋Ao HMGS、Ao FPPS和Ao SS的表达与总皂苷及薯蓣皂苷元的含量呈明显负相关,而同一模式、同一嫩茎不同部位间并不存在相关关系,其可能与留母茎采收过程中,母茎中合成薯蓣皂苷元及总皂苷能部分转运至商品笋中有关。该研究确定留母茎芦笋采收具有较好的皂苷含量,是值得推广的生产模式。

摘  要:
To study the effects of different asparagus harvesting mode on yield,saponin contents and genes' expression of saponin biosynthesis,by using three year-age asparagus cultivar Jersey Knight variety which were planned at asparagus base in Mile City of Yunnan as materials,in this study,3 harvesting models,eg. white spears collection,bald green spears collection and green spears collection with mother stem were performed in 100 m2 experiment plots respectively in same tunnel green house.The yields were calculated,the contents of total saponins and diosgenin,an intermediate of asparagus saponins biosynthesis were determined. Based on asparagus EST data,primers were designed to ampli-fied full coding sequence( CDS) of hydroxymethyl coenzyme A synthetase( Ao HMGS),farnesene synthase( Ao FPPS) and squalene synthase( Ao SS) which were catalyze the key steps in early saponin biosynthesis pathway,and their expression were detected by method of q-RT-PCR amplification. The results showed that spear colleting with mother fern get the,highest contends of total saponins and diosgenin,while white spear collecting showed lowest yield and contents of diosgenin but total saponin was higher than green spear colleting with bald way. The q-RT-PCR amplification showed that the expression of Ao HMGS,Ao FPPS and Ao SS genes and contents of total saponin and diosgenin presented a significant negative correlation among different harvesting mode,while there didn't exist correlation in different parts of the same spears cultivated in same mode. The possible reason is that the saponins and diosgenin synthesized in stalks are partially transferred to spears under the stalk maintained collection mode. It seems that the collecting spears with mother fern is the best way to get high quality of asparagus,even this mode needs more labors and times for spear colleting.
