摘  要:
多糖是家蚕幼虫体内富含的活性成分之一,为了建立高效的蚕体水溶性粗多糖提取工艺,以家蚕5龄第3天幼虫为材料提取水溶性粗多糖。在单因素试验的基础上,采用Box-Behnken设计对提取温度、提取时间和原料质量浓度3个因素进行中心组合试验,研究各因素交互作用及对蚕体水溶性粗多糖提取率的影响,采用Design-Expert软件拟合得到预测提取率的二次回归方程。试验结果表明,提取时间和原料质量浓度的交互作用对蚕体水溶性粗多糖提取率具有极显著影响,3个因素对提取率贡献大小的排序为原料质量浓度>提取时间>提取温度。运用响应面法优化得到最优提取工艺条件为:提取温度80℃,提取时间97 min,原料质量浓度37.5 g/L。优化的提取工艺简单、稳定,蚕体水溶性粗多糖的实际提取率为3.042%,与预测值(3.055%)非常接近,具有应用价值。

摘  要:
Polysaccharide is one of the active ingredients rich in silkworm( Bombyx mori) larval body. In order to establish efficient extraction process of water-soluble crude polysaccharide from silkworm larval body,the 3rd day silkworm larva of the 5th instar was used as material to extract water-soluble crude polysaccharide. Based on single-factor experiments,three factors including extraction temperature,extraction time and material mass concentration were selected to conduct the central composite experiment using Box-Behnken design method. Interaction of these factors and their influence on the yield of water-soluble crude polysaccharide form silkworm larval body were studied,and a quadratic regression equation was established through Design-Expert software. The results showed that interaction of extraction time and material mass concentration had extremely significant effect on the yield of water-soluble crude polysaccharide from silkworm larval body. The ranking of three factors was material mass concentration > extraction time > extraction temperature,according to their contribution to the yield of water-soluble crude polysaccharide from silkworm larval body. Combined with response surface methodology,the optimal process condition for the extraction was determined as follows:extraction temperature 80 ℃,extraction time 97 min and material mass concentration 37. 5 g / L. The optimized extraction process had the characteristics of simple and stable.Under this condition,the practical yield of water-soluble crude polysaccharide in extract was 3. 042%,being very close to the predicted value 3. 055%. Thus,the optimized extraction process has practical value.
