摘 要:
在充分了解SP2/0细胞系的生长、增殖和遗传特性后,以聚乙二醇(PEG)诱导动物细胞融合为契机,意在探索不同分子量、不同浓度的PEG作融合剂对诱导SP2/0细胞与脾细胞融合的最适条件,在HAT选择培养基下通过细胞融合率的变化进行比较。结果表明,分子量为4000,浓度为50%的PEG诱导的细胞融合率最高,为下一步制备狂犬病毒疫苗单克隆抗体奠定基础。

摘 要:
Based on a comprehensive understanding of the property of the growth,proliferation and genetic characteristics of SP2/0 cell line,animal cell fusion method was performed to explore the optimal condition for the fusion between SP2/0 cell and spleen cells under different molecular weight and gradient of PEG as fusion agent.The comparison was carried out by the variation of the cell fusion in HAT selection medium.The conclusion indicated that the cell fusion yield was much higher when the concentration of PEG was 50% and the molecular weight was 4 000,which provides a basis for the preparation of monoclonal antibodies in next step.
