聚乙烯亚胺配合灭活禽流感病毒口服免疫对鸡免疫反应和生长发育的影响

摘  要:
[目的]本文旨在研究聚乙烯亚胺(PEI)配合灭活H5N1禽流感病毒口服免疫对鸡免疫反应和生长发育的影响。[方法]1日龄雏鸡35只,随机分成5组,每组7只。根据免疫方法分为口服生理盐水0.3 m L组(CK)、口服灭活禽流感病毒0.25 m L组(IAIV)、口服0.08 g聚乙烯亚胺配合0.25 m L灭活禽流感病毒组[IAIV+PEI(0.08)]、口服0.02 g聚乙烯亚胺配合0.25 m L灭活禽流感病毒组[IAIV+PEI(0.02)]和皮下注射灭活禽流感油乳佐剂疫苗组(i H)。分别在首次免疫后第3、5、7周通过检测口腔棉拭子、肠道和气管涮洗液中特异性Ig A、血清中特异性Ig G和HI抗体水平对免疫效果进行评价,然后称体质量,检测空肠和回肠绒毛长度以对生长发育效果进行评价。[结果]0.02 g聚乙烯亚胺配合0.25 m L灭活禽流感病毒口服免疫雏鸡的口腔棉拭子和肠道涮洗液中特异性Ig A水平显著高于单独口服灭活病毒雏鸡组(P<0.05);0.08 g和0.02 g聚乙烯亚胺分别配合0.25 m L灭活禽流感病毒口服免疫雏鸡后第7周,血清中特异性Ig G水平极显著升高(P<0.01);0.02 g聚乙烯亚胺配合0.25 m L灭活禽流感病毒口服免疫雏鸡后,其空肠、回肠绒毛长度比单独口服灭活病毒极显著增长(P<0.01)。[结论]0.02 g聚乙烯亚胺配合0.25 m L灭活H5N1禽流感病毒口服免疫雏鸡,不仅增强免疫反应,提高免疫力,还能促进营养物质吸收。

摘  要:
[Objectives]An experiment was conducted to investigate the effect of oral immunization with H5N1 inactivated Avian influenza virus(IAIV) and adjuvant polyethyleneimine(PEI) on the immune response and growing development in chicken.[Methods]According to the immune method,35 one day old chickens were randomly divided into 5 groups,oral immunization with normal saline 0.3 m L(CK),oral immunization with IAIV 0.25 m L(IAIV),oral immunization with 0.08 g PEI plus IAIV 0.25 m L[IAIV+PEI(0.08) ],oral immunization with 0.02 g PEI plus IAIV 0.25 m L[IAIV+PEI(0.02) ],subcutaneous injection with inactivated oil-adjuvant emulsion vaccine(i H).3,5,7 weeks after the first vaccination,antigen-specific mucosal Ig A titers in mouth swab,tracheal and intestinal wash,the antigen-specific serum Ig G titers and HI titers were detected to evaluate the immune effect.Weight and the length of intestinal villus in jejunum and ileum were detected to evaluate growing development.[Results]The antigen-specific Ig A antibody titers in mouth swab and intestinal wash induced by 0.02 g PEI plus 0.25 m L IAIV were substantially(P<0.05) greater than antigen-alone treatment.7 weeks after the first vaccination,the level of Ig G in serum induced by serum induced by 0.08 g PEI and 0.02 g PEI plus 0.25 m L IAIV were more significantly increased(P<0.01) than antigen-alone treatment.The length of intestinal villus in jejunum and ileum of 0.02 g PEI group were more significantly(P < 0.01) increased than antigen-alone treatment.[Conclusions]Chicken oral immunization with 0.02 g PEI and 0.25 m L H5N1 IAIV could strengthen immune response and absorption of nutrients.
