奶牛源乳房链球菌毒力与耐药性分析

摘  要:
乳房链球菌是导致奶牛乳房炎的主要病原菌之一,但近年来研究发现乳房链球菌也与奶牛子宫内的感染相关,导致子宫内膜炎的发生。采集甘肃省不同地区5个牧场患有子宫内膜炎奶牛的子宫黏液样品48份,共计分离得到18株乳房链球菌。然后通过CAMP试验来检测分离菌株的协同溶血作用,采用普通PCR检测10种已知毒力基因(hasA、hasB、hasC、sua、slp、pauA、gapC、acdA、fpb、cfu)的携带率,通过确定8种抗菌药物(氨苄西林、红霉素、四环素、左氧氟沙星、利福平、克林霉素、头孢噻呋、庆大霉素)的最小抑菌浓度(MIC)来了解乳房链球菌的耐药性,以及利用结晶紫染色法对乳房链球菌形成的生物被膜进行定量检测。结果表明,所有分离株均携带pauA、fpb、sua、slp、gapC及cfu基因,而基因hasA、hasB、acdA的携带率分别为88.89%、94.44%和94.44%,未检测到hasC基因。此外,CAMP试验阳性菌为7株(38.89%)。18株乳房链球菌都对利福平敏感,11株(61.1%)、14株(77.8%)、14株(77.8%)分别对红霉素、左氧氟沙星、氨苄西林敏感,对四环素、克林霉素、头孢噻呋及庆大霉素耐药菌株的比例分别为77.8%、77.8%、66.7%和72.2%。所有的乳房链球菌分离株都具有形成生物被膜的能力,其中6株(33.3%)具有较强的生物被膜形成能力。研究结果对了解乳房链球菌对子宫内膜炎的致病机制,监测病原菌耐药情况,以此来指导临床合理用药,并对甘肃省奶牛子宫内膜炎的综合防控提供理论依据。

摘  要:
Streptococcus uberis is the main pathogen leading to cow mastitis.Futhernore,in recent years,studies found that S.uberis was also related to the infection of uterine leading to the endometritis.In this study,we recovered 18 S.uberi strains from 48 uterus mucus samples in 5 herds in Gansu province.Then we did CAMP test,detected 10 virulence genes(hasA,hasB,hasC,sua,slp,pauA,gapC,acdA,fpb,cfu)by ordinary PCR,antimicrobial resistance(ampicillin,erythromycin,tetracycline,levofloxacin,rifampicin,clindamycin,ceftiofur,and gentamycin)by microbroth dilution method to determine MIC value,and quantitatively detected the biofilm formation abiliby by crystal violet staining method.The results showed that all the isolates carried the pauA,fpb,sua,slp,gapCand cfu genes.The positive rates of hasA,hasBand acdAwere 88.89%,94.44%and 94.44%,respectively,and no hasCgene was detected in this study.In addition,7 strains(38.89%)were showed CAMP test positive.All 18 isolates were sensitive to rifampicin.11 strains(61.1%),14 strains(77.8%),14 strains(77.8%)were sensitive to erythromycin,levofloxacin,ampicillin.And the proportions of resistance to tetracycline,clindamycin,ceftiofur and gentamicin were 77.8%,77.8%,66.7%,72.2%,respectively.All isolates had the ability to form biofilm and 6 isolates(33.3%)had strong ability to form biofilm.In this study,we monitored the drug resistance and could guide clinical antimicrobial treatment of cow endometritis.Furthermore,we try to figure out the pathogenic mechanism of S.uberis and provide theoretical basis for comprehensive prevention and control of cow endometritis in Gansu province.
