不同小麦品种幼胚愈伤诱导及适宜草甘膦浓度的筛选

摘  要:
以6个小麦品种(郑麦9023、郑麦366、周麦18、矮抗58、花培5号、花培8号)幼胚为外植体,研究了不同基因型间幼胚愈伤组织诱导、分化成苗以及对草甘膦敏感性的差异。结果表明,品种间出愈时间和初始分化时间有明显差异,花培5号和花培8号的出愈和初始分化时间最短,成苗率较高,可作为优良的受体材料。设置0,1,2,3,4 mmol/L5种草甘膦浓度研究不同小麦品种对草甘膦的敏感性,结果显示,小麦幼胚愈伤分化期与诱导筛选阶段相比,褐化死亡较明显,是理想的筛选时期。草甘膦浓度为3 mmol/L时,郑麦9023、周麦18、郑麦366的分化率与对照相比呈现显著差异,而与4 mmol/L时这3个品种的分化率无显著差异。矮抗58在4种草甘膦筛选浓度下与对照相比分化率逐渐降低,在4 mmol/L时分化率为2.08%,大部分的愈伤生长受到抑制。花培5号和花培8号与其他几个品种相比,对草甘膦更加敏感,筛选浓度为2 mmol/L时与对照相比分化率显著降低,分别为3.42%和4.17%。因此,在愈伤分化时期用草甘膦筛选时,郑麦9023、郑麦366、周麦18适宜的筛选浓度为3 mmol/L,矮抗58为4 mmol/L,花培5号和花培8号为2 mmol/L。

摘  要:
In this paper,we studied differences of immature embryo calli,proliferation,differentiation and glyphosate sensitivity in different genotypes by using the immature embryo of six wheat( Zhengmai 9023,Zhengmai 366,Zhoumai18,Aikang 58,Huapei NO. 5 and Huapei NO. 8) cultivars as explants. The results showed that the callus formation time and initial time of differentiation had a significant difference in different wheat cultivars,Huapei NO. 5 and Huapei NO. 8 were optimal for tissue culture because the index of callus formation time and initial time were higher than others,regenerated plantlet yield reached high level. Screening of wheat varieties to glyphosate sensitivity through 0,1,2,3,4 mmol / L 5 kinds of glyphosate concentration,results showed differentiation and Callus Browning death compared to more obvious,is the ideal screening period. The concentration of 3 mmol / L Zhengmai 9023,Zhoumai 18,Zhengmai 366 differentiation rate compared with the control group showed significant differences,but no significant difference with 4 mmol / L. Aikang 58 in the screening concentration of glyphosate compared with control differentiation rate decreased gradually,in 4 mmol / L of glyphosate screening differentiation rate reached 2. 08%,most of the callus growth was inhibited. For Huapei No. 5,Huapei No. 8 and other varieties compared tomore sensitive to glyphosate,screening concentration was 2 mmol / L,when compared with the control differentiation rate decreased,3. 42% and 4. 17%,respectively.
