黄芪发酵菌的筛选鉴定及纤维素酶学性质试验

摘  要:
为分离发酵黄芪菌株,试验采取黄芪样品接种营养琼脂,置30℃环境培养。取分离菌接种纤维素刚果红琼脂,筛选能形成较大降解圈的细菌进行鉴定,并测定其纤维素酶性质。结果表明,筛选菌为解淀粉芽孢杆菌,该菌株降解圈直径(H)与菌落直径(C)的比值(H/C)为4.03。在37℃环境培养36 h酶活力达到高峰,为32.16 U/m L。在低于60℃及pH值6.0~8.0环境纤维素酶较稳定,可保持酶活力90%以上。

摘  要:
In order to isolate the strains which have fermenting performance for Radix Astragalus,the Radix Astragali samples were inoculated on nutrient agar medium and cultivation at 30 ℃,and then the isolating strains were inoculated on the cellulose-congo red agar,Screening the bacteria which can form a larger cellulose-decomposing zone to identification based on molecular biological method was performed,and the activity of the cellulose was determined. The results showed that the isolated strain was identified as Bacillus amyloliq-uefaciens,and the ratio of H/C was 4. 03. The highest cellulase activity reached 32. 16 U/m L when cultivated at37 ℃ for 36 h. The cellulase activity was stable at temperatures lower than 60 ℃ and at pH range of 6. 0-8. 0,which retained more than 90%.
