摘  要:
用所建立的反相高效液相色谱-荧光检测法测定干酪中的生物胺。采用的色谱柱为HP Hypersil ODS柱(125mm×4.6mm×5μm);荧光检测激发波长为340nm,发射波长为450nm;洗脱程序为梯度洗脱,流速为1.0ml/min。干酪样品经邻苯二甲醛柱前自动衍生,能使组胺、酪胺、腐胺、尸胺、色胺在15min内得到良好分离,并且各生物胺浓度在0.01~25mg/L时其浓度与相应峰面积有良好的线性关系,最低检测限(以信噪比为3计)为0.005~0.01mg/L。样品加标各生物胺的回收率为92.6%~107.4%。该法快速准确、简便灵敏、分离度高,能够满足干酪中生物胺的检测要求。

摘  要:
A high-performance liquid chromatographic(HPLC)procedure for determining 5 biogenic amines was introduced. The method was based on a reversed-phase column(HP Hypersil ODS column,125mm×4.6 mm×5μm)for precolumn derivatization with o-phthaldialdehyde and fluorometric detection(excitation wave length at 340nm and emission wave length at 450 nm).Biogenic amines are separated by gradient solution,with the flow rate 1.0 ml/min.It allowes simultaneous determination of 5 amines in less than 15 min for histamine and tyramine,putrescine,cadaverine and tryptamine.Linearity for each amine is observed between 0.01 to 25 mg/L.Detection limits range from 0.005 to 0.01 mg/L.The average recovery of cheese for biogenic amines was 92.6%~107.4%.The method is simple,fast and reliable.It can meet the determination needed for biogenic amines in cheeses.
