超声联合酶法提取平菇多糖工艺优化

摘  要:
以平菇子实体为原料,采用超声波联合酶法,通过单因素及响应面试验设计对平菇多糖提取工艺进行优化优化,考察液固比、酶解温度、酶解pH值和超声时间四个因素对平菇多糖提取率的影响.经研究得出,平菇多糖最优提取工艺为:液固比22∶1、每处理温度60℃、酶解pH7.0、超声时间65 min,在此工艺条件下平菇提取率85.68%,理论预测值为85.73%,相对误差0.06%,理论值与实际值相近,说明该工艺可行,希望对平菇多糖标准化生产提供参考.

摘  要:
The single factor test was used to determine that the enzyme mass fraction(1.5%)and the enzymatic hydrolysis time(60 min)were the best in the extraction test of Pleurotus ostreatus polysaccharides. On this basis,the effects of liquid-solid ratio,enzyme treatment temperature,enzymatic pH value and ultrasonic treatment time on the extraction rate of Pleurotus ostreatus polysaccharides were studied by four-factor quadratic regression orthogonal rotation combination design method,and the optimum extraction process of Pleurotus ostreatus polysaccharides was determined. The results showed that the optimum extraction process of Pleurotus ostreatus polysaccharides was as follows:the ratio of liquid to solid was 22 ∶1,the treatment temperature was 60 ℃,the pH of enzymatic hydrolysis was 7.0,and the ultrasonic time was 65 min. Under this condition,the theoretical prediction value was 85.73% and the relative error was 0.06%.
