阿魏侧耳胞外多糖生物活性初步研究

摘  要:
本文研究了阿魏侧耳液体发酵胞外多糖的生物活性。液体深层发酵和乙醇萃取法制备阿魏侧耳胞外多糖,小鼠经灌胃给药,采用血液分析仪、体外孵育等方法,测定血液中白细胞数量、腹腔巨噬细胞吞噬能力和免疫器官指数;利用显微观察、细胞计数和纸片琼脂法等研究阿魏侧耳胞外多糖对肿瘤细胞、细菌和真菌生长的影响。结果表明:与对照组相比,灌胃100和200 mg/kg的阿魏侧耳胞外多糖可显著提高小鼠血液白细胞数量(P<0.05)、增加免疫器官指数(P<0.05)和增强巨噬细胞吞噬能力(P<0.05);40~80 g/L阿魏侧耳胞外多糖对枯草芽孢杆菌的生长有一定的抑制作用;5~80 mg/L阿魏侧耳胞外多糖对体外培养的人胃癌细胞和人宫颈癌细胞的存活无明显影响作用。因此,一定浓度阿魏侧耳胞外多糖可提高小鼠的免疫能力,抑制枯草芽孢杆菌的生长,但对其他供试细菌、真菌和体外培养的肿瘤细胞的生长无明显影响作用。

摘  要:
In this paper,the bioactivity of extracellular polysaccharides produced by Pleurotus ferulae Lanzi was studied. Extracellular polysaccharides,which were ethanolic extracts,were prepared from submerged fermentation culture medium of Pleurotus ferulae Lanzi and intragastrically administered to mice. The number of leukocytes in blood,peritoneal macrophage phagocytic activity and weight index of immune organ were examined with automatic blood analyzer and incubation in vitro,et al. Effects of extracellular polysaccharides produced by Pleurotus ferulae Lanzi on tumor cells,bacteria and fungi were examined with microscope,cell counting or the agar diffusion method. The results showed that extracellular polysaccharides produced by Pleurotus ferulae Lanzi at the concentration of 100 and 200 mg /kg obviously increased the number of leukocytes in blood( P < 0. 05) and the weight index of spleen and thymus gland( P < 0. 05) and enhanced the phagocytic activities of murine peritoneal macrophage( P < 0. 05). Extracellular polysaccharides in concentration range of 40- 80 g /L could inhibit the proliferation capacity of Bacillus subtilis,but extracellular polysaccharides in concentration range of 5- 80 mg /L had no influence on the survival of MGC80-3 and Hela229. It was concluded that extracellular polysaccharides produced by Pleurotus ferulae Lanzi in certain concentration range could enhance the immune function of mice and inhibit the proliferation capacity of Bacillus subtilis but had no effect on the growth of the other tested bacteria,fungi or tumor cells cultured in vitro.
