黄羽肉鸡日粮中添加胆汁酸盐对肠道黏膜转运蛋白及消化酶的影响

摘  要:
试验旨在研究日粮中添加不同水平的胆汁酸盐对黄羽肉鸡雏鸡肠道黏膜转运蛋白(SREBF1、FATP4)及消化酶(FAS、ACC)基因表达的影响。将420只雏鸡分为7组,试验1组为阴性对照组饲喂基础日粮,试验2组为阳性对照组,在基础日粮上添加0.02%泰乐菌素代替麸皮,试验3组、试验4组、试验5组、试验6组、试验7组分别用0.015%、0.025%、0.035%、0.045%、0.055%的胆汁酸盐代替麸皮。雏鸡饲养28 d,在28日龄时采集十二指肠、空肠、回肠和肝脏,通过实时荧光定量PCR技术对各组织相应目的基因进行表达量测定。结果显示:黄羽肉鸡不同组织SREBF1、FATP4、FAS、ACC mRNA的相对表达量均存在极显著差异(P<0.01);不同胆汁酸盐添加水平黄羽肉鸡肠道SREBF1、FATP4、FAS、ACC mRNA相对表达量存在极显著差异(P<0.01)。由此可见,黄羽肉鸡肠道营养转运蛋白SREBF1、FATP4及消化酶FAS、ACC基因表达具有肠段及胆汁酸盐添加水平的差异性。日粮添加0.055%胆汁酸盐,对于肠道整体营养转运蛋白及消化酶的基因表达影响最佳。

摘  要:
In this reserch, the effect of adding different levels of bile salts in the diet on the genetic expression of intestinal mucosal transport proteins(SREBF1, FATP4) and digestive enzymes(FAS, ACC) in yellow-feather broiler chicks was studied. A total of 420 chicks and breeders were divided into seven groups. The chickens in test group 1(negative control group) were fed with the basal diet, and the chickens in test group 2(positive control group) were fed the basal diet supplemented with 0.02% tyloxin instead of bran. Bran in groups 3, 4, 5, 6 and 7 were substituted with 0.015%, 0.025%, 0.035%, 0.045%and 0.055% of bile salts, respectively. The chickens were fed with for 28 days, and the duodenum, jejunum, ileum and liver were collected at the age of 28 days and the expression levels of the corresponding target genes in each tissue were determined by real-time quantitative fluorescence qutea PCR. The relative expression levels of SREBF1, FATP4, FAS, and ACC mRNA in different tissues of yellow feather broiler chicks were extremely different(P<0.01); the relative expression levels of SREBF1, FATP4,FAS, ACC mRNA in the intestine of yellow feather broilers chicks were extremely different(P<0.01). It could be seen that the expression of intestinal nutrient transporter SREBF1 and FATP4 and the digestive enzymes FAS and ACC of yellow-feather broilers chicks changed with the level of bile salts supplemental. Adding 0.055% bile salts in the diet had the best effect on the gene expression of the whole nutrient translocator and digestive enzyme in the intestine.
