SYBR Green I实时定量PCR方法检测2型猪圆环病毒

摘  要:
利用PCR技术扩增出2型猪圆环病毒(PCV)ORF2保守区基因107 bp片段,并克隆到pMD-19-T载体上,纯化的质粒作为PCR检测的标准模板,以10倍梯度稀释质粒为标准模板,进行荧光定量PCR,并绘制标准曲线。再以提取的PCV-2、PCV-1、猪细小病毒(PPV)和猪伪狂犬病毒(PRV)的DNA作为模板,采用同样体系进行实时PCR检测。结果表明:建立的实时定量PCR方法,起始模板浓度与样本阈值循环数(Ct)值之间呈很好的线性关系,标准曲线斜率为-0.32,相关系数r2为0.997,Ct值为11.9~26.9。PCV-2能扩出S型荧光曲线,熔解曲线的峰狭窄单一,而PCV-1、PPV和PRV均不能扩出。对32份健康猪和32份断奶仔猪多系统消耗综合征(PMWS)可疑猪样品定量检测:健康猪阳性率为44%(14/32),病毒载量均小于104拷贝.μL-1;PMWS可疑猪阳性率96.8%(31/32),病毒载量均大于105拷贝.μL-1。实时荧光定量PCR方法可用于2型猪圆环病毒定性及定量的检测。

摘  要:
Using PCR assay,a 107 bp region of the porcine circovirus type 2(PCV-2) ORF2 gene was cloned to pMD-19-T vector,and the constructed plasmid was named pT-PCV-2 107.Serial dilutions of plasmid pT-PCV-2 107 were used to quantify the virus genomic copy number,and served as standards curve.PCV-2,PPV,and PRV were detected at the same time with the Realtime PCR.The results showed that the regression coefficient of the quantitative curve was 0.997 and the Tm of melting curve analysis was(81.53±0.16) ℃.By comparison of viral load in sample from 32 healthy animals and 32 PMWS animals,healthy pigs had viral load less than 104 copies per μL and 44%(14/32) were tested positive for PCV-2,while all clinically sick PMWS pigs had PCV-2 loads above 105 copies per μL and 96.8%(31/32) were tested positive for PCV-2.Real time PCR assay can be used to monitor and detect PCV-2 with SYBR Green I.
