利用甘薯淀粉生产废水培养侧孢短芽孢杆菌的研究

摘  要:
为探明利用甘薯淀粉生产废水培养侧孢短芽孢杆菌(Brevibacillus laterosporus)的最适条件,采用单因素试验,研究了甘薯淀粉废水浓度、初始p H值、培养温度对侧孢短芽孢杆菌菌体生长的影响。结果表明:甘薯淀粉废水浓度40%,初始p H值为7.0~8.0,培养温度为30~35°C,以2%比例接种侧孢短芽孢杆菌种子液,150r·min-1培养24h时,菌体浓度最高可达1.63×109cfu·m L-1。在此基础上,应用Placket-Burman设计,对微生物培养基中常用的8种无机盐离子(KH2PO4,Mg SO4,Ca Cl2,Na Cl,Fe SO4,Cu SO4,Mn SO4和Zn SO4)进行了评价。结果表明:向甘薯淀粉废水中添加不同种类或不同浓度的外源无机盐离子,对菌体生长的影响不显著。试验结果初步证明了利用甘薯淀粉生产废水发酵培养侧孢短芽孢杆菌的可行性,为甘薯淀粉生产废水的资源化利用和低成本生产侧孢短芽孢杆菌提供了一条可参考的技术路线。

摘  要:
This experiment was conducted to explore the optimum culture conditions of Brevibacillus laterosporus using sweet potato wastewater. The effects of concentration, initial p H and temperature on the production of B. laterosporus were investigated by single-factor experiments. With SPSW diluted to 40%, initial p H 7.0-8.0, temperature 30-35°C, inoculum 2%, rotate speed at150 r·min-1, a maximum biomass of 1.63×109cfu·m L-1was achieved after 24 h cultivation. The two-level Placket-Burman(PB)design was used to explore the effects of mineral salts(KH2PO4, Mg SO4, Ca Cl2, Na Cl, Fe SO4, Cu SO4, Mn SO4, Zn SO4) that generally used as components of microbial medium. The effects on the production of B. laterosporus with different kinds or concentrations of mineral salts added to SPSW were not significant. Our experiments proved the feasibility to culture B.laterosporus with SPSW as low-cost medium, and provided an environmental friendly way to utilize SPSW.
