胶质瘤中survivin蛋白的表达及其生物学意义

摘  要:
目的:探讨survivin蛋白在胶质瘤中的表达情况及其与增殖指数、凋亡指数和微血管密度的相关性。方法:采用S-P法检测胶质瘤56例和正常脑组织8例中survivin蛋白、Ki67和FⅧRAg的表达,肿瘤细胞的凋亡采用TUNEL检测,并结合病理特点进行分析。结果:Ⅰ~Ⅱ、Ⅲ、Ⅳ级胶质瘤中,sur-vivin蛋白的表达率分别为25.3%、56.8%、83.2%,组织学分级之间差异有显著性(P<0.05);survivin表达阳性者的Ki67、微血管密度明显高于survivin阴性者,而其凋亡率则低于survivin阴性者,差异有显著性(P<0.05);正常脑组织中未见survivin表达。结论:Survivin在胶质瘤中高表达,与其恶性程度、凋亡、增殖活性及血管生成密切相关。

摘  要:
Objective: To explore the relationships of survivin expression with proliferation, apoptosis and angiogenesis of gliomas. Methods: The expressions of survivin, Ki67 and Factor Ⅷ-related antigen (FⅧRag) were detected by immunohistochemistry and Apoptotic cells of tumors were screened by TdT-mediated dUTP nick end-labeling(TUNEL) in the paraffin-embeded sections of 56 gliomas and 8 cases normal brain tissue. Results: Expression of survivin was significantly associated with malignant grade of gliomas.Furthermore, survivin-positive gliomas had higher Ki67 labeling index(LI) and MVD than that of survivin-negative gliomas(P<0.05),and AI was inversely correlated with survivin expression.No survivin expression was detected in the tissue samples from normal brain.Conclusion: In gliomas, survivin overexpression has an extremely close relationship to malignant proliferation, antiapoptosis and angiogenesis of brain gliomas.
