利巴韦林对感染猫细小病毒细胞的治疗作用研究

摘  要:
为了研究利巴韦林对感染猫细小病毒(FPV)细胞的作用,试验采用F81猫肾细胞,在猫细小病毒感染后不同时间点收集细胞,应用MTT法检测细胞的增殖活性;利用利巴韦林毒性试验,摸索出药物对细胞的安全浓度;利用不同浓度利巴韦林处理已攻毒细胞,MTT法检测细胞的增殖活性,酶标法检测细胞氧化损伤情况。结果表明:利巴韦林能明显抑制FPV引起的细胞死亡率,病毒组过氧化氢(H2O2)含量和细胞培养液中乳酸脱氢酶(LDH)活性在各个采样时间点均显著高于对照组,显示细胞氧化损伤严重,致使LDH从细胞内渗出。病毒组细胞内超氧化物歧化酶(SOD)活性和还原型谷胱甘肽(GSH)含量均明显低于对照组,揭示病毒所引起的细胞氧化损伤进一步激发了细胞自体保护性ROS清除机制,最终导致了细胞内SOD活性和GSH含量的下降。利巴韦林药物组LDH活性和H2O2含量都呈现升高趋势,但其增长幅度明显低于病毒组;在各采样时间点,药物组细胞内SOD活性和GSH含量均明显(P<0.05)高于病毒组,低于对照组。说明利巴韦林对感染猫细小病毒细胞具有一定的修复治疗效果。

摘  要:
To study the effect of ribavirin on the cells infected with Feline parvovirus( FPV),the feline kidney( F81) cells infected with FPV at different time points were collected to detect the activity of the cell proliferation using MTT assay. The toxicity test of ribavirin was performed to confirm the safe concentration to the cells. The challenged cells were treated with different concentrations of ribavirin,and the MTT assay and the enzyme- linked immunosorbnent assay were used to detect the proliferation activity and oxidative damage of the cells,respectively. The result showed that ribavirin could inhibit the death rate of the cells caused by FPV,the content of hydrogen peroxide and lactate dehydrogenase( LDH) activity in the cell culture fluid significantly increased in the FPV infection group compared with the control group at each time point,showing serious oxidative damage of the cells and resulting in LDH leaking from the cells. The superoxide dismutase( SOD) activity and the content of reduced glutathione( GSH) obviously decreased in the FPV infection group compared with the control group,revealing that the oxidative damage of the cells caused by FPV could further stimulate ROS clearance mechanisms of autologous protection for the cells,resulting in the decreases of SOD activity and the content of GSH. The LDH activity and the content of hydrogen peroxide were showing an increasing trend in the ribavirin treatment group,but its growth rate was obviously lower than that in the FPV infection group; while SOD activity and the content of GSH in the ribavirin treatment group was noticeably higher( P < 0. 05) than that in the FPV infection group and lower than that in the control group. The results indicate that ribavirin has certain restoration and treatment effects on the cells infected with FPV.
