鸡枞菌多糖对小鼠T细胞免疫功能的影响

摘  要:
采用鸡枞菌多糖(TAP)体外作用于小鼠T淋巴细胞,用ELISA法检测其IL-4、IFN-γ及IL-2水平,流式细胞仪检测T淋巴细胞亚群,RT-PCR法检测相关细胞因子mRNA表达量,观察研究了鸡枞菌多糖对小鼠T细胞免疫功能的影响。结果表明,与ConA组相比,25~100μg/mL TAP在作用72h内均能显著促进T细胞产生IL-4、IFN-γ及IL-2(P<0.01);25μg/mL TAP作用48h后可显著降低被ConA激活的T细胞CD4+%(P<0.01),升高CD8+%(P<0.05),并降低CD4+/CD8+(P<0.05);50μg/mL TAP则可显著升高CD3+%(P<0.01);50μg/mL TAP在作用72h内能显著提高IL-2、IL-4及IFN-γmRNA表达量(P<0.01),并使其保持较高水平。提示,TAP对细胞免疫功能具有显著增强作用,T淋巴细胞是TAP的靶细胞之一。

摘  要:
To observe the influence of Termitomyces albuminosus polysaccharide(TAP)on T cell immunity in mouse,splenic lymphocyte of mouse was exposed to TAP in vitro.IL-4,IFN-γ and IL-2 were detected by ELISA,T lymphocyte subpopulations were detected by flow cytometry,and the expression of related cytokines was detected by RT-PCR.The results showed that,in comparison with ConA group,25 to 100 μg/mL TAP could significantly increase level of IL-4,IFN-γ and IL-2 in 72 h(P<0.01),and after treatment with 25 μg/mL TAP in 48 h,CD4+ % of T cell(P<0.01) and CD4+/CD8+(P<0.05) were all depressed,but CD8+ % was raised(P<0.05);CD3+ % was raised by 50 μg/mL TAP(P<0.05).IL-4,IFN-γ and IL-2 mRNA expression significantly increased by 50 μg/mL TAP,and they were kept with higher expression level than those of ConA group during 72 h(P<0.01).In conclusion,TAP could notablely enhance cellular immune function,and T lymphocyte was the target cell of TAP.
