也门铁嵌合体品种在离体培养中的变异规律及调控

摘  要:
以金心也门铁Dracaena fragrans cv. massangeana及金边也门铁D. fragrans cv. lindenii为材料,对其叶片嵌合性状在离体培养过程中的变异规律及通过调节培养条件降低其变异率进行了研究.在离体培养过程中,金心也门铁和金边也门铁的嵌合体性状极不稳定,后代中分别有4类和8类变异,2个品种之间也可相互变异;变异率高低主要受芽增殖方式的影响,由愈伤组织分化出的不定芽的变异率高达70%以上,由丛生芽基部组织分化出的不定芽的变异率约40%~50%,而由腋芽分化方式产生的芽变异率只有13%~17%,但后者的增殖率太低.将基部带有分生组织的丛生芽(增殖10代)在MS+1.0 mg/LBA+0.1 mg/LIAA+3 mg/LAgNO3上增殖培养后,2品种的变异率分别能降为22.80%和25.34%,有效增殖率比以腋芽增殖方式提高约1倍.

摘  要:
Variation of leave chimera character during in vitro culture and its variation rate were studied by using Dracaena fragrans cv.massangeana and D.fragrans cv.lindenii as the material.In vitro culture,the characters of chimera in massangeana and lindenii were extremely unstable,there were four categories and 8 kinds of variation respectively in descendants and the mutual variation between the two species;their variation rates were mainly affected by the regeneration bud way,the mutation rate of adventitious buds was over 70% from callus tissue differentiation,approximately 40% to 50% from numerous basal bud differentiation,and only 13% to 17% from the axillary bud differentiation.After numerous basal buds multiplied 10 generations cultured on MS+1.0 mg/L BA + 0.1 mg/L IAA+ 3 mg/L AgNO3, the variation rate of two varieties reduced to 22.80% and 25.34%,and effective multiplication rate of numerous basal buds was about 2 times higher than that of the axillary buds.
