天山雪莲叶片全长cDNA文库的构建

摘  要：
以天山雪莲(Saussurea involucrataKar.et Kir)幼嫩叶片为研究材料,利用CreatorTMSMARTTMcD-NA Library Construction技术构建了天山雪莲叶片全长cDNA文库,原始文库滴度达3.93×105cfu/mL,文库插入片段多在1 000 bp左右。随机挑取10个克隆进行测序,通过生物信息学初步分析表明,100%为全长cDNA,大部分可以在GenBank中找到同源序列。天山雪莲叶片全长cDNA文库的构建为雪莲基因资源保存和植物抗寒机理的研究奠定了基础。

摘  要：
A full-length cDNA Library from the Saussurea involucrata Kar.et Kir laminae was constructed by using CreatorTM SMARTTM cDNA Library Construction Protocol.The titer of cDNA library was estimated as 3.93×10~5 cfu/mL,which was about 500 times coverage of gene expressional profile,and the inserted fragments in the library are about 1000 bp length.Ten clones were randomly selected to sequence and bioinformatics analysis was carried out,all of the sequences were full-length cDNA,and most of the sequences were homologous to the genes known in GenBank.This full-length cDNA Library provides a important tool for the study of the cold resistence mechanisms of the Saussurea involucrata Kar.et Kir.
