摘 要:
Tn3是存在于生物体内的一种非常重要的转座酶,其是基因组中具有一段特异的转位特性的独立的DNA序列,从而对目的基因起调控作用。转座酶Tn3具有基因敲除的功能,Gal4能识别DNA启动子上游两端保守的17bp长的一段序列,因此将Tn3与Gal4相组合,实现靶向敲除目的基因的目的。论文综述了转座酶Tn3的结构及其转座机制、Gal4的结构及其机制、Tn3-Gal4系统的简介和酶分子的定向进化,旨在为今后靶向基因的敲除以及靶向基因的修复提供研究思路。

摘 要:
Tn3 is a very important transposase present in organisms,which enables independent DNA sequences with a specific translocation characteristic in the genome to regulate the gene of interest.Studies have shown that the transposase Tn3 has a knock-out function,and Gal4 recognizes a conserved sequence of17 bp long upstream of the DNA promoter,thus combining Tn3 with Gal4 to achieve the purpose of targeting the knockout target gene.This paper reviewed the structure of transposase Tn3 and its transposition mechanism,the structure and mechanism of Gal4,the introduction of Tn3-Gal4 system and the directed evolution of enzyme molecules.It aimed to provide research ideas for the knockout of targeted genes and the repair of targeted genes in the future.
