酵母硒对铅暴露致蛋鸡卵巢毒性损伤的保护效应研究

摘  要:
本试验旨在探讨铅(Pb)暴露对产蛋鸡卵巢的毒性损伤及酵母硒的保护效应。将90只40周龄海蓝褐蛋鸡随机分成3组,每组30只,每组3个重复,每个重复10只。采用饮水染毒方式,对蛋鸡进行铅暴露(100 mg/L)和酵母硒保护试验(饲料中添加3%酵母硒),试验周期8周。结果表明,与对照组比较,Pb染毒组鸡血清E2、P4含量呈类似变化,自第4周显著下降(P<0.05);卵巢组织中SOD、GSH-Px活性和GSH含量均显著或极显著降低(P<0.05或P<0.01),MDA含量极显著升高(P<0.01);卵巢铅含量极显著升高(P<0.01)。超微结构观察发现,Pb染毒组卵巢颗粒细胞染色质边集,线粒体嵴断裂,空泡化,内质网肿胀扩张,溶酶体集聚;卵母细胞微绒毛断裂。与Pb染毒组比较,Se保护组血清E2、P4含量自第6周显著升高,卵巢SOD活性、GSH含量显著升高(P<0.05),MDA含量显著降低(P<0.05);卵巢硒含量极显著升高,铅含量显著下降,线粒体、细胞核、内质网、微绒毛等损伤均明显减轻。表明铅引起的脂质过氧化损伤是铅致蛋鸡卵巢损伤的重要原因,氧化应激引起的卵巢颗粒细胞内质网损伤和细胞凋亡可能是造成血清雌激素水平下降的原因之一。酵母硒通过减少铅蓄积,缓解卵巢组织脂质过氧化损伤,对蛋鸡铅暴露引起的卵巢损伤具有一定的保护作用。

摘  要:
This study aimed to determine the effects of dietary supplementation with yeast selenium on lead(Pb) toxicity on ovaries in laying hens.A total of 90 40-week-old hyline hens were randomly divided into three groups.Thrity birds in each group,three repeats in each group,10 birds in each repeat.Lead acetate(100mg Pb/L in water) and/or yeast selenium(3% in feed) were administered for eight weeks.The results showed that the content of E2 and P4 in serum in group Pb showed a similar change compared with the control,all decreased signifi cantly from the fouth week(P<0.05).Activity of total SOD,GSH-Px and content of GSH in ovary significantly decreased(P<0.01),while MDA increased very significantly(P<0.01).Content of lead in ovary increased very significantly(P<0.01).Ultrastructurally,nucleoli damaged with crush and salvation,mitochondria damaged with swelling,disappearance and fragmentation of carina and microvilli following the exposure of lead were observed.Compare with group Pb,the content of E2 and P4in group Se increased significently from the sixth week.Both activity of total SOD and content of GSH increased significantly(P<0.01).While,MDA decreased significently(P<0.05).The content of selenium increased while lead decreased significantly(P<0.05).There also no obvious changes in Ultrastructure.In conclusion,lipid peroxidation is the most important reason for lead toxicity,apoptosis of granular cell and damage of endoplasmic reticulum may cause the decrease of hormone in serum in laying hens.Yeast selenium protects ovary by reducing accumulation of lead and alleviating oxidative stress caused by lead.
